摘 要:为了了解麝鼠Klf5基因结构、生物学功能及在麝鼠香腺中的表达差异,探究Klf5基因在香腺发育过程中的作用,试验自2019年开始于2月末选购约10月龄、体况相近的麝鼠若干只,于2—9月份每月月末各随机挑选3只,适应性饲养2 d后取两侧香腺,进行RNA提取与反转录并对Klf5基因进行测序,对Klf5基因编码序列(CDS)的编码蛋白进行生物信息学分析,并利用实时荧光定量PCR(qPCR)技术分析成年雄性麝鼠繁殖周期内香腺组织中Klf5基因转录水平的差异表达,同时探讨不同物种间Klf5基因的进化关系。结果表明:麝鼠Klf5基因CDS全长1 353 bp,可编码450个氨基酸,为不稳定的、亲水性、非分泌、非跨膜蛋白,主要定位于细胞核,有3个典型的锌指结构域。Klf5基因在麝鼠香腺中相对表达量自2—9月份整体呈上调—下调—上调—下调的双峰表达趋势。麝鼠Klf5基因与仓鼠科缓行田鼠、草原田鼠和水鼠平遗传距离最近。说明在麝鼠香腺发育和泌香周期变化过程中,Klf5基因与麝鼠香分泌相关并参与香腺细胞发育调控,在进化上较为保守。In order to understand the structure and biological function of muskrat Klf5 gene, its differential expression in the muskrat scent gland, and to explore the role of Klf5 in the development process of the scent gland, the experiment purchased several 10-month-old muskrats with similar body condition in late February 2019. Three muskrats were randomly selected each month from February to September, and the scent glands were taken from both sides after 2 d of acclimatization. RNA extraction and reverse transcription were performed and the Klf5 gene was sequenced, and bioinformatic analysis was performed on the encoded protein of the coding sequence(CDS) of the Klf5 gene. Real-time quantitative gene amplification fluorescence assay technique was used to analyze the differential expression of Klf5 gene transcriptional levels in the scent gland tissues during the reproductive cycle of adult male muskrats, and the evolutionary relationship of Klf5 gene among different species was also discussed. The results showed that the CDS of muskrat Klf5 gene was 1,353 bp long and could encode 450 amino acids, which was an unstable, hydrophilic, non-secretory, non-transmembrane protein mainly localized in the nucleus with three typical zinc finger structural domains. The expression of Klf5 gene in muskrat scent gland showed an overall bimodal expression trend of up-down-up-down regulation from February to September. The muskrat Klf5 gene was genetically closest to those of creeping voles, prairie voles, and water voles in the Cricetidae family. The results suggested that during the development of muskrat scent gland and scent secretion cycle changes, the Klf5 gene was in relation to muskrat scent secretion and involved in the regulation of scent gland cell development, which was evolutionarily conserved.
