基于16S rRNA测序分析巴马小型猪性别与肠道微生物的关系

摘  要:为了研究性别对巴马小型猪粪便微生物组成及丰度的影响,试验以10头雄性和10头雌性试验用巴马小型猪作为试验对象,收集新鲜粪便,采用16S rRNA测序技术和生物信息学分析方法,分析微生物多样性、组成并对预测的代谢通路进行了差异分析。结果表明:雄性巴马小型猪和雌性巴马小型猪之间微生物的Alpha多样性无显著差异(P>0.05)。在门水平上雄性和雌性巴马小型猪的核心菌群均为厚壁菌门(Firmicutes)和拟杆菌门(Bacteroidetes);在属水平上雄性的核心菌群为志贺氏杆菌属(Shigella,5.81%)、颤螺菌属(Oscillospira,4.62%)、密螺旋体属(Treponema,4.14%)、梭菌属(Clostridiaceae_Clostridium,2.73%)及拟杆菌属(Bacteroides,2.58%),雌性为颤螺菌属(5.23%)、密螺旋体属(4.94%)、链球菌属(Streptococcus,3.74%)、SMB53(2.35%)及梭菌属(2.12%)。在门水平上,雌性疣微菌门(Verrucomicrobia)的丰度显著高于雄性(P<0.05);在属水平上,雄性链霉菌属(Streptomyces)、双歧杆菌属(Bifidobacterium)及拟杆菌属的丰度显著高于雌性(P<0.05),雌性链球菌属、多尔氏菌属(Dorea)、考拉杆菌属(Phascolarctobacterium)、阿克曼氏菌属(Akkermansia)、L7A_E11及p_75_a5的丰度显著高于雄性(P<0.05)。此外,未发现雌性和雄性之间有显著差异的代谢通路。说明雄性和雌性巴马小型猪粪便微生物的Alpha多样性无显著差异,但在门和属水平上部分菌群的丰度存在显著差异。In order to investigate the effects of gender on the composition and abundance of fecal microorganisms in Bama Miniature pigs, the experiment was conducted with 10 male and 10 female experimental Bama Miniature pigs as test subjects, and fresh feces were collected;16 S rRNA sequencing technology and bioinformatics analysis were used to analyze microbial diversity, composition and functional prediction. The results showed that there was no significant difference in microbial Alpha diversity between male and female Bama Miniature pigs(P>0.05). At the phylum level, the core microfloras of both male and female Bama Miniature pigs were Firmicutes and Bacteroidota. At the genus level, the core microfloras of males were Shigella 5.81%, Oscillospira 4.62%, Treponema 4.14%, Clostridium 2.73%, and Bacteroides 2.58%;those of females were Oscillospira 5.23%, Treponema 4.94%, Streptococcus 3.74%, SMB53 2.35%, and Clostridium 2.12%. LEfSe(LDA Effect Size) analysis showed that the abundance of Verrucomicrobia was significantly higher in females than in males(P<0.05) at the phylum level. At the genus level, the abundance of Streptomyces, Bifidobacterium and Bacteroides in males was significantly higher than those in females(P<0.05);the abundance of Streptococcus, Dorea, Phascolarctobacterium, Akkermansia, L7 A_E11 and p_75_a5 in females were significantly higher than those in males(P<0.05). In addition, during the differential analysis of predicted metabolic pathway, no metabolic pathways were found to be significantly different between females and males. The results suggested that there was no significant difference in the Alpha diversity of fecal microorganisms of male and female Bama Miniature pigs, but there were significant differences in the abundance of some microfloras at the phylum and genus levels.
