摘  要:为初步了解河北邢台及周边地区猪繁殖与呼吸综合征病毒(PRRSV)流行情况,本试验于2021年3—10月在邢台及周边地区采集217份疑似PRRSV感染病料,应用荧光定量PCR (qPCR)技术检测PRRSV流行情况,同时对部分类NADC30毒株ORF5基因序列进行扩增和分析。结果显示,该地区PRRSV平均检出率为48.39%(105/217),不同地区检出率介于29.85%~75.0%;进一步基因分型发现,类NADC30毒株、HP-PRRSV毒株和经典型PRRSV毒株所占比例分别为60.95%、31.43%和9.52%;8个毒株ORF5基因序列均与类NADC30毒株HeN1同源性最高,为99.0%~99.5%,与其他毒株对应序列同源性相对较低;进一步遗传进化分析结果显示,8个毒株与类NADC30毒株聚为同一分支。结果表明,PRRSV在河北邢台及周边地区流行情况十分严重,其中类NADC30毒株为优势流行株,提示猪场应更加注重生物安全防控,加强对PRRSV流行及变异情况的监测工作。In order to assess the prevalence of porcine reproductive and respiratory syndrome virus(PRRSV) in Xingtai and its surrounding areas, this study collected 217 tissue samples suspected with PRRS infection from these areas during 2021.3 to 2021.10. The prevalence of PRRSV was determined by fluorescence quantitative PCR(qPCR), and the open reading frame 5(ORF5) gene sequences of eight NADC30-like PRRSV strains were amplified and analyzed. The results showed that the average detection rate of PRRSV was 48.39%(105/217),and the detection rates in different areas varied from 29.85% to 75.0%. The proportions of NADC30-like PRRSV,highly pathogenic PRRSV(HP-PRRSV) and classical PRRSV strains were 60.95%,31.43% and 9.52%,respectively. The ORF5 gene sequences of eight PRRSV strains revealed the highest homology(99.0%~99.5%) with NADC30-like PRRSV strain HeN1,and displayed relatively low homologies with other strains. Further genetic analysis showed that eight strains and NADC30-like PRRSV strains from GenBank clustered into the same branch. Thus, PRRSV is prevalent in Xingtai city and its surrounding areas of Hebei Province, and NADC30-like PRRSV strain appears to dominate among the strains, highlighting the need for pig farmers to increase the biosafety level and continuously monitor the prevalence and genetic variation of PRRSV.
