甜菜碱缓解皮质酮诱导的鸡肝脏胆固醇沉积的分子机制研究  被引量:2

摘  要:本试验主要探究母鸡饲喂甜菜碱能否缓解皮质酮诱导的子代鸡肝脏胆固醇沉积及其分子机制。120只如皋黄产蛋鸡随机分为空白组和甜菜碱组,空白组母鸡饲喂基础饲粮,甜菜碱组母鸡饲喂含0.5%甜菜碱的饲粮,饲喂28 d后收集受精蛋进行孵化;出壳小鸡饲养至49日龄时,按照平均体重分别从空白组中挑选16只和甜菜碱组中挑选8只雄性鸡,空白组的16只鸡再按照体重随机分成A和B 2组,其中空白A组鸡注射等体积的生理盐水(对照组),空白B组(皮质酮组)和甜菜碱组鸡(甜菜碱+皮质酮组)按4 mg/kg体重的剂量皮下注射皮质酮,连续注射7 d。结果显示,与对照组相比,皮质酮处理显著提高(P <0.05)鸡肝脏中胆固醇的含量;甜菜碱可有效降低皮质酮诱导的肝脏胆固醇沉积。进一步研究发现,甜菜碱有效缓解(P <0.05)皮质酮处理造成子代鸡肝脏胆固醇合成基因SREBP1和HMGCR mRNA和蛋白表达的升高。此外,胆固醇降解基因CYP27A1在皮质酮处理后显著下降(P <0.05),而甜菜碱没有缓解效应(P> 0.05)。以上研究结果表明,母鸡饲喂甜菜碱可能通过抑制子代雄性鸡肝脏中胆固醇合成基因SREBP1和MHGCR基因的表达来缓解皮质酮诱导的肝脏胆固醇沉积。The purpose of this study was to investigate whether maternal betaine supplementation could alleviate corticosterone(CORT)-induced hepatic cholesterol accumulation in offspring chickens and its molecular mechanism.One hundred and twenty Rugao yellow-feathered laying hens were randomly divided into normal and betaine groups.Hens were fed basal diet or 0.5%betaine-supplemented diet for 28 days before the eggs were collected for incubation.The newly hatched chicks were raised at 49 d of age,16 male chickens from normal group and 8 male chickens from betaine group were selected,and 16 male chickens in the normal group were divided into two subgroups(A and B),the chickens in normal A group were subjected to saline(Control),and the chickens in the normal B(CORT)and betaine groups(betaine+CORT)were subjected to CORT(4.0 mg/kg body)treatment with daily subcutaneous injection for 7 days.The results showed that CORT treatment significantly increased(P < 0.05)the chicken hepatic cholesterol content compared to the control;while prenatal betaine exposure prevented CORT-induced increase(P < 0.05)in hepatic cholesterol content.Further study revealed that prenatal betaine exposure prevented CORT-induced upregulation of heaptic mRNA and protein expression of cholesterol biosynthesis genes SREBP1 and HMGCR in offspring chickens.Besides,betaine-treated chickens were not able to protect from CORT-induced repression(P < 0.05)in CYP27A1 expression and betaine had no alleviating effect(P > 0.05).These results suggest that maternal betaine supplementation alleviates CORT-induced hepatic cholesterol deposition most probably through inhibiting SREBP1 and HMGCR genes expression in offspring chickens.
