摘 要:为了解Cofilin在鸭肠炎病毒(DEV)感染过程中的作用及DEV致病机制。以受精鸭蛋、鸭肠炎病毒GZ株为主要研究对象,根据测序获得鸭源-Cofilin2全基因序列,提取和筛选Cofilin2-shRAN,利用FQ-PCR对细胞Cofilin基因转录水平进行测定,分析Cofilin基因的RNAi对DEV的影响。DEF细胞转染处理过程中,转染成功的干扰质粒共有4个,其中Cofilin2-shRAN-86沉默效益最高,可促进DEV的增殖效果。To understand the role of Cofilin in the process of duck enteritis virus(DEV)infection and the pathogenic mechanism of DEV.With fertilized duck eggs and duck enteritis virus GZ as the main research object,the complete sequence of duck derived Cofilin 2 gene was obtained according to sequencing,Cofilin2 shRAN was extracted and screened,and the transcription level of Cofilin gene in cells was measured by FQ-PCR to analyze the effect of RNAi of Cofilin gene on DEV.In the process of DEF cell transfection,there are 4 interfering plasmids successfully transfected,among which Cofilin2-shRAN-86 has the highest silencing effect and can promote the proliferation of DEV.
