摘 要:目的:探讨蛹虫草多糖对高脂高胆固醇诱导ob/ob小鼠非酒精性脂肪肝的影响。方法:40只雄性ob/ob小鼠随机分为模型组、非诺贝特组[20 mg/(kg·d)]及[25、50、100 mg/(kg·d)]蛹虫草多糖组,均以高脂高胆固醇饮食(脂肪34.90%、胆固醇22.80%)饲养。12周末,酶法检测血清TC、TG、HDL-C水平,赖氏法检测肝组织中ALT与AST活力;分别采用WST-1法和TBA法检测肝组织SOD活性与MDA含量;分别采用酶法和ELISA方法测定肝组织TG、TC、FFAs含量;HE染色检测肝组织脂质蓄积水平。结果:与模型组相比,100 mg/(kg·d)蛹虫草多糖下调小鼠血浆TC和TG含量;3个剂量蛹虫草多糖组均不同程度升高HDL-C水平;升高肝组织SOD活性,下调肝组织MDA含量;50、100 mg/(kg·d)剂量蛹虫草多糖下调肝组织TG和TC含量,降低ALT活性,100 mg/kg·d蛹虫草多糖下降AST活性;HE染色结果显示蛹虫草多糖显著改善肝脏脂质变性。结论:泰山蛹虫草多糖可通过调控血脂、减轻氧化应激水平改善高脂高胆固醇诱导的脂肪肝病变,对非酒精性脂肪肝具有良好防治效果。Objective:To explore the effects of Cordyceps Militaris Polysaccharide from Taishan on fatty liver phenotype in ob/ob mice which induced by high-fat and high-choleeterol diet.Methods:40 male ob/ob mice were randomly divided into model group,Fenofibrate group and three doses Polysaccharide groups.Mice were all given high-fat and high-cholesterol diet.The levels of TC,TG and HDL-C in serum were detected by enzymatic method.The activities of ALT and AST were determined by Reich method.The activity of SOD and the content of MDA were tested with WST-1 method and TBA method respectively.The contens of TC,TG and FFAs in liver were detected by enzymatic method and ELISA methods,steatosis of liver tissue were tested by HE staining.Results:In contrast to the models,the levels of TC and TG of serum were reduced in 100 mg/(kg·d)Polysaccharide group.The levels of HDL-C in serum were all increased in three dose Polysaccharide groups,respectively.Meanwhile,the activities of SOD were all increased,and the contents of MDA were declined.The levels of TG and TC were reduced in 50 mg/(kg·d)and 100 mg/(kg·d)groups,and the activities of ALT were decreased.The activities of AST were lessened in 100 mg/(kg·d)group,while it was no significant change in other two groups.The HE staining experiments showed that Cordyceps Militaris Polysaccharide could improve steatosis of liver tissue and enhance liver function.Conclusion:The Cordyceps Militaris Polysaccharide from Taishan could prevent and treat nonalcoholic fatty liver disease via reducing lipid and improving fatty liver phenotype by regulating lipids and reducing oxidative stress level.It could prevent and cure nonalcoholic fatty liver disease.
