玉屏风散和芪寄方对流感病毒裂解疫苗注射小鼠免疫反应的影响

摘　　要：目的探讨玉屏风散和芪寄方对流感病毒裂解疫苗保护效力的影响。方法40只Balb/c小鼠随机分为空白组、免疫对照组、铝佐剂组、玉屏风散组、芪寄方组,每组8只。玉屏风散组小鼠给予玉屏风散0.25 g/10 g灌胃,芪寄方组小鼠给予芪寄方0.25 g/10 g灌胃,其余各组小鼠给予纯净水0.25 ml灌胃。各组均每日灌胃1次,连续10天。给药4天,空白组小鼠经腿部肌肉注射0.1 ml PBS,其余各组小鼠经腿部肌肉注射季节性流感病毒裂解疫苗(IVV)0.75μg进行初次免疫,其中铝佐剂组IVV含100μg氢氧化铝。在初次免疫2周后各组以同样方法加强免疫1次。每隔7天称取小鼠体质量1次,观察小鼠一般情况,至给药28天。采用ELISA法检测初次免疫后7、14、21、28天血清特异性抗体IgG水平,初次免疫后28天检测血清特异性抗体IgG亚型IgG_(1)、IgG_(2)a的水平;留取脾脏称重,计算脾脏指数,并采用流式细胞术检测脾脏记忆性B细胞比例。结果各组间小鼠同时间体质量差异无统计学意义(P>0.05)。与本组给药前及给药7天时比较,芪寄方组小鼠给药14天时体质量明显下降(P<0.05)。与本组给药14天时比较,玉屏风组和芪寄方组给药28天时体质量明显上升(P<0.05)。各组小鼠毛发柔顺光滑,一般活动正常,排泄物质地无明显异常。与空白组和免疫对照组相比,芪寄方组小鼠脾脏指数、记忆性B细胞比例明显升高(P<0.05或P<0.01)。与空白组比较,各组小鼠血清IgG_(1)和IgG_(2)a水平,各时间点IgG水平均升高(P<0.05或P<0.01)。与免疫对照组相比,玉屏风散组初次免疫后21、28天,芪寄方组初次免疫后28天IgG水平均显著升高(P<0.05或P<0.01);与铝佐剂组相比,玉屏风散组初次免疫后21天IgG水平显著升高(P<0.05)。与免疫对照组相比,玉屏风散组和芪寄方组IgG_(1)水平,铝佐剂组IgG_(2)a水平均显著升高(P<0.05或P<0.01);与铝佐剂组相比,玉屏风散组和芪寄方组IgG_(1)水显著Objective To explore the effect of oral administration of Yupingfeng Powder(玉屏风散)and Qiji Formula(芪寄方)on the protective efficacy of the split virus influenza vaccine.Methods Forty Balb/c mice were randomly divided into blank group,immune control group,aluminum adjuvant group,Yupingfeng Powder group and Qiji Formula group,with eight mice in each group.Yupingfeng Powder group and Qiji Formula group were given 0.25 g/10 g of corresponding formulas by gavage,while the other groups were given 0.25 ml purified water by gavage,once every day for 10 days.After 4-day medication,mice in the blank group were injected intramuscularly with 0.1 ml of PBS on the legs,while those in other groups were injected with 0.75μg of seasonal influenza virus split vaccine(IVV)for initial immunization,and additionally 100μg of aluminum hydroxide was contained in IVV in the aluminum adjuvant group.Two weeks after the initial immunization,the mice in all groups were continued for a booster injection.The mice were weighed every 7 days to observe the general condition.ELISA method was used to detect the level of serum specific antibody IgG 7,14,21 and 28 days after the initial immunization,and 28 days after the initial immunization,the level of serum specific antibody subtype IgG_(1)and IgG_(2)a was tested,the spleen was weighed to calculate the spleen index.The proportion of memory B cells in the spleen was detected by flow cytometry.Results There was no significant differences in the body weight of the mice among the groups regarding the same measurement time(P>0.05).The body weight significantly decreased after 14-day medication in the Qiji Formula group compared to that measured before and after 7-day medication(P<0.05).Compared to those measured after 14-day medication,the body weight after 28-day medication significantly increased in the Yupingfeng Powder group and Qiji Formula group(P<0.05).The hair of the mice in all groups was soft and smooth,with normal general activities and no obvious abnormality in the excrement.Co
