发酵黄芪对肉鸡脾脏指数及淋巴细胞增殖的影响

摘　　要：为了研究发酵黄芪对肉鸡脾脏免疫功能的影响,试验选取1日龄健康雄性817肉鸡120只,利用单因子完全随机法分为空白对照组及试验Ⅰ、Ⅱ、Ⅲ组,每组3个重复,每个重复10只鸡。空白对照组肉鸡饮用自来水,不添加任何药物或添加剂,试验Ⅰ、Ⅱ、Ⅲ组分别在自来水中添加0.5‰植物乳酸菌、0.5‰黄芪水提物、0.5‰发酵黄芪水提物,试验周期为21 d。第21天时采集各组肉鸡脾脏,称重并测定脾脏指数,制备脾脏石蜡切片,分别经苏木精-伊红染色和兔源增殖细胞核抗原(PCNA)免疫组织化学染色检测脾脏组织形态结构变化及脾脏淋巴细胞增殖情况。结果表明:与空白对照组相比,试验Ⅱ、Ⅲ组脾脏指数分别增加了11.41%和15.44%,试验Ⅰ组脾脏指数却有所下降,均差异不显著(P>0.05)。试验Ⅰ、Ⅱ、Ⅲ组鸡只的脾脏颜色、外观及组织结构与空白对照组相比均无明显变化;与空白对照组相比,试验Ⅲ组脾脏淋巴细胞数增加了33.53%,差异显著(P<0.05)。说明发酵黄芪能促进肉鸡脾脏生长发育和淋巴细胞增殖,改善脾脏的免疫功能,提高鸡只免疫力,但具体机制还需要深入研究。In order to study the effects of fermented Astragalus on spleen immune function in broilers,in this experiment,1201-day-old 817 healthy male broilers were selected and divided into blank control group,test groups Ⅰ,Ⅱ and Ⅲ using the one-way complete randomization method,with three replicates of 10 chickens in each group.The broilers in the blank control group drank tap water without adding any drugs or additives,while 0.5‰ plant Lactobacillus,0.5‰Astragalus aqueous extract and 0.5‰ fermented Astragalus aqueous extract were added to tap water in test groups Ⅰ,Ⅱ and Ⅲ,respectively,for a period of 21 d.On day 21,the spleens of broilers in each group were collected,weighed and the spleen indexes were measured;and the morphological structure changes of the spleen tissue and the proliferation of spleen lymphocytes were detected by paraffin section-hematoxylin eosin staining and immunohistochemical staining of rabbit-derived proliferating cell nuclear antigen(PCNA),respectively.The results showed that compared with the blank control group,the spleen indexes increased by 11.41% and 15.44% in groups Ⅱ and Ⅲ,respectively,but decreased in group Ⅰ;all differences were not significant(P>0.05).There were no significant changes in color,appearance and tissue structure of the spleen of broilers between blank control group and test groups Ⅰ,Ⅱ,Ⅲ.Compared with the control group,the spleen lymphocyte count increased by 33.53% in group Ⅲ,which was significant(P<0.05).The results indicated that fermented Astragalus could promote the growth and development of broiler spleen and lymphocyte proliferation,which could improve the immune function of spleen and enhance the immunity of chickens,but the specific mechanism needed to be studied in depth.
