摘  要:目的探讨芪锦颗粒对被动型Heymann肾炎大鼠的肾保护作用及其机制。方法采用尾静脉一次性注射抗肾小管刷状缘提取物(FxlA)抗体构建被动型Heymann肾炎大鼠模型,将造模成功大鼠随机分为模型组,醋酸地塞米松组(0.07 mg/kg),芪锦颗粒低、中、高剂量组(2、4、8 g/kg),另设正常组,每组12只,灌胃给予相应药物,每天1次,连续4周。第0、7、35天检测各组大鼠24 h尿蛋白定量(UPQ)及血清肌酐(Cr)、尿素氮(BUN)水平,HE染色观察肾组织病理学变化,免疫荧光染色观察肾组织中免疫球蛋白(IgG)沉积情况,Western blot法检测肾组织NF-κB p65、NF-κB抑制蛋白α(IκBα)及环氧化酶(COX-2)蛋白表达。结果与正常组比较,模型组大鼠肾组织损伤较严重,第7、35天UPQ、Cr、BUN水平升高(P<0.01),肾组织中IgG沉积以及NF-κB p65、COX-2蛋白表达升高(P<0.01),而IκBα蛋白表达降低(P<0.01);与模型组比较,醋酸地塞米松组和芪锦颗粒中、高剂量组大鼠肾组织损伤明显减轻,第35天UPQ、Cr、BUN水平降低(P<0.05),肾组织中IgG沉积以及NF-κB p65、COX-2蛋白表达降低(P<0.05),IκBα蛋白表达升高(P<0.05),而芪锦颗粒低剂量组各指标无明显变化(P>0.05)。结论芪锦颗粒可改善被动型Heymann肾炎大鼠的肾损伤,其机制可能与抑制NF-κB信号通路活化有关。AIM To investigate the renal protective effect of Qijin Granules on passive Heymann nephritis(PHN)rats and its mechanism.METHODS The successfully established PHN rat models by one injection of anti-renal tubular brush edge extract(FxlA)antibody into tail vein were randomly divided into model group,dexamethasone acetate group(0.07 mg/kg),Qijin Granules groups for low dose,medium dose and high dose interventions(2,4,8 g/kg)respectively,with 12 rats in each group,in contrast to another intact rats in the normal group.The corresponding drugs were dosed upon the rats once daily by gavage.The levels of urinary protein quantification(UPQ),serum creatinine(Cr)and urea nitrogen(BUN)were measured on days of 0,7 and 35.The renal histopathological changes were observed by HE staining;the immunoglobulin(IgG)deposition assessed by immunofluorescence staining;and the protein expressions of NF-κB p65,NF-κB inhibitory proteinα(IκBα)and cyclooxygenase-2(COX-2)detected by Western blot.RESULTS Compared with the normal group,the model group demonstrated seriously damaged renal tissue,increased levels of UPQ,Cr and BUN on the 7th and 35th day(P<0.01),increased IgG deposition and NF-κB p65 and COX-2 protein expressions(P<0.01),and decreased IκBαprotein expression(P<0.01).Compared with the model group,dexamethasone acetate group and Qijin Granules group intervened with medium and high doses shared significantly reduced renal tissue injury,decreased levels of UPQ,Cr and BUN on the 35th day(P<0.05),decreased renal IgG deposition and NF-κB p65 and COX-2 protein expressions(P<0.05),and increased IκBαprotein expression(P<0.05),but low dose Qijin Granules group was found with no effect in terms of all indices levels assessment(P>0.05).CONCLUSION Qijin Granules improve the kidney injury of PHN rats,and the mechanism may be associated with the inhibition of NF-κB signal pathway activation.
