摘 要:目的研究大青叶Folium isatidis Fort.中生物碱类成分。方法大青叶75%乙醇提取液正丁醇萃取部位采用大孔树脂、硅胶、ODS、Sephadex LH-20以及制备液相色谱进行分离纯化,根据理化性质及波谱数据鉴定化合物的结构。结果从中分离得到17个生物碱,分别鉴定为5-羟基-2-吲哚酮(1)、3-hydroxy-3-(2-hydroxyethyl)indolin-2-one(2)、烟酰胺(3)、2′-甲氧基尿苷(4)、safowerine A(5)、胸苷(6)、次黄嘌呤(7)、indoline-2-one-5-O-β-D-glucopyranoside(8)、腺苷(9)、吲哚-3-甲酸-6-O-β-D-葡萄糖苷(10)、transtorine(11)、苯丙氨酸(12)、左旋色氨酸(13)、anoectochine(14)、吲哚-3-乙腈(15)、isatindigotindoline A(16)、色胺酮(17)。结论化合物2~6、8、10~11、13~14为首次从菘蓝属植物和该植物中分离得到。AIM To study the alkaloid constituents from Isatis indigotica Fort..METHODS The n-butanol fraction of 75%ethanol extract from I.indigotica was isolated and purified by column chromatography over macroporous resin,silica,ODS,Sephadex LH-20 and preparative liquid chromatography,then the structures of obtained compounds were identified by physicochemical properties and spectral data.RESULTS Seventeen alkaloid compounds were isolated and identified as 5-hydroxy-2-indolinone(1),3-hydroxy-3-(2-hydroxyethyl)indolin-2-one(2),nicotinamide(3),2′-O-methyluridine(4),safowerine A(5),thymidine(6),hypoxanthine(7),indoline-2-one-5-O-β-D-glucopyranoside(8),adenosine(9),6-hydroxylindole-3-carboxylic acid-6-O-β-D-glucopyranoside(10),transtorine(11),phenylalanine(12),L-tryptophan(13),anoectochine(14),indole-3-acetonitrile(15),isatindigotindoline A(16),tryptanthrin(17).CONCLUSION Compounds 2-6,8,10-11,13-14 were isolated from I.indigotica and this plant for the first time.
