摘  要:目的:优选南板蓝根中多糖的提取工艺,建立其含量测定的方法。方法:以葡萄糖为对照品,采用苯酚-硫酸法测定多糖的含量。以多糖提取率作为评价指标,考察提取方法、料液比、提取时间、醇浓度对南板蓝根中多糖提取工艺的影响。结果:多糖提取条件为30倍水加热回流提取50 min,以85%的乙醇沉淀多糖为佳。葡萄糖的质量浓度在10.20~203.9μg/mL范围内与吸光度呈良好的线性关系(r^(2)=0.9991),平均回收率为101.25%(RSD为2.43%)。15批南板蓝根样品多糖含量在35.23~58.13 mg/g之间。结论:优选的提取工艺稳定可行,该方法操作简单、重复性好,可为南板蓝根中多糖的含量测定提供理论依据。We optimized the extraction process of polysaccharides from Baphicacanthus cusia,and established a method for the determination of its content.The content of polysaccharides was determined by phenol-sulfuric acid method with glucose as the reference.The effect of extraction method,solid-liquid ratio,extraction time and alcohol concentration on the extraction process of polysaccharides from Baphicacanthus cusia was investigated by using the extraction rate of polysaccharides as the evaluation index.The optimum extraction process was heating reflux extraction for 50 min with 30 times water,and precipitating with 85%ethanol.The linear relationship between the absorbance and the glucose concentration was good in the range of 10.20μg/mL to 203.9μg/mL(R^(2)=0.9991).The average recovery was 101.25%(RSD=2.43%).The content of polysaccharides in 15 batches of Baphicacanthus cusia samples was between 35.23 mg/g and 58.13 mg/g.The optimized extraction process is stable and feasible.The method is simple and reproducible,and can provide theoretical basis for the determination of polysaccharides in Baphicacanthus cusia.
