摘　　要：目的:研究松节方醇提物对类风湿性关节炎大鼠模型T细胞抗原受体(TCR)Vβ亚族基因表达谱的影响。方法:采用类风湿性关节炎(RA)大鼠模型,给予松节方2 w,观察RA大鼠踝关节组织病理学变化,免疫组化检测RA大鼠踝关节滑膜组织中TCR表达情况,提取大鼠血液、滑膜组织总RNA,qRT-PCR法分析Tcr Vb亚族基因变化。结果:与正常对照组相比,模型对照组大鼠足关节肿胀百分率、关节指数及组织半定量评分明显升高,关节滑膜组织TCR强阳性表达,同时血液Tcr Vb1、Vb5的mRNA表达明显减弱,Vb6 mRNA表达明显增强(P<0.05或P<0.01);与模型对照组相比,松节方醇提物能明显抑制RA大鼠滑膜组织炎性细胞浸润和滑膜组织增生,下调滑膜组织TCR阳性表达,同时上调血液Tcr Vb1、Vb3、Vb5及滑膜组织Tcr Vb1、Vb2、Vb5、Vb11、Vb12 mRNA表达,下调血液Tcr Vb4、Vb6、Vb7及滑膜组织Tcr Vb7 mRNA表达。结论:松节方醇提物对RA大鼠的作用机制可能是调节Tcr Vb的表达,改善免疫识别,激活有效免疫反应来对抗外来抗原。Objective:To study the effect of ethanol extract of Songjie Formula(松节方)(SJF)on the gene expression profiling of T cell receptor(TCR)Vβsubgroup in rat model of rheumatoid arthritis(RA).Methods:RA rats were treated with SJF(2 weeks).The histopathological changes of ankle joint were observed and the expression of TCR in synovial tissues of the ankle joint in RA rats was detected by immunohistochemistry.Total RNA was extracted from blood and synovial tissues of RA rats and the expression of TCR Vβsubgroup genes was analyzed by qRT-PCR.Results:Compared with the normal control group,the model group had high foot joint swelling percentage,joint index,and tissue semi-quantitative score,strong positive expression of TCR in joint synovial tissues,low mRNA expression of TCR Vβ1 and Vβ5,and high mRNA expression of Vβ6(P<0.05 or P<0.01).Compared with the model group,ethanol extract of SJF significantly inhibited the infiltration of inflammatory cells and proliferation of synovial tissues and down-regulated the positive expression of TCR in synovial tissues of RA rats.The ethanol extract of SJF up-regulated the mRNA expression of TCR Vβ1,Vβ3,and Vβ5 in blood and mRNA expression of TCR Vβ1,Vβ2,Vβ5,Vβ11,and Vβ12 in synovial tissues and down-regulated the gene expression of TCR Vβ4,Vβ6,and Vβ7 in blood and TCR Vβ7 in synovial tissues.Conclusion:The potential mechanism of ethanol extract of SJF against RA in rats is that it regulates TCR Vβexpression,improves immune recognition,and activates effective immune response against external antigens.
