摘  要:目的:清热解毒软胶囊由川芎和当归挥发油组成,本文探讨清热解毒软胶囊防治流感病毒、脂多糖(LPS)所引起的急性肺损伤相关机制,为后期开发奠定坚实的基础。方法:采用静脉注射LPS和气管局部滴注流感病毒建立急性肺损伤小鼠模型,考察灌胃清热解毒软胶囊对急性肺损伤小鼠的保护作用,同时利用皮下注射角叉菜胶诱导小鼠尾部血栓模型,考察清热解毒软胶囊改善微循环的作用。结果:与模型对照组比较,清热解毒软胶囊2、4、8 g/kg可明显延长静脉注射LPS 20 mg/kg小鼠或H1N1流感病毒感染小鼠的生存时间,降低其死亡率(P<0.05或P<0.01);与正常对照组比较,模型对照组肺指数及肺组织病理评分显著升高,肺组织中还原型谷胱甘肽(GSH)的含量降低,丙二醛的含量显著升高(P<0.01);防治性给药7次后,清热解毒软胶囊可显著改善上述指标,尤其是清热解毒软胶囊8 g/kg组(P<0.05或P<0.01)。与正常对照组比较,模型对照组皮下注射角叉菜胶致小鼠尾部血栓形成长度显著增加(P<0.01),凝血酶时间缩短和纤维蛋白原含量升高(P<0.05或P<0.01);预防性给药5次后,清热解毒软胶囊1、2、4 g/kg组可明显改善上述指标(P<0.05或P<0.01)。结论:清热解毒软胶囊能有效延长流感病毒感染小鼠、大剂量静脉注射LPS小鼠的生存时间,减少其死亡率;其对急性肺损伤的保护作用可能与减轻肺部氧化应激、减少血栓形成、减轻肺水肿有关。Objective:Qingrejiedu(清热解毒)Soft Capsules are composed of volatile oils of Chuanxiong(川芎)and Danggui(当归).This paper aimed to explore the underlying mechanism of Qingrejiedu Soft Capsules in the prevention and treatment of acute lung injury induced by influenza virus and lipopolysaccharide(LPS)to lay a solid foundation for further development.Methods:The acute lung injury model was established by intravenous injection of LPS and local instillation of influenza virus into the trachea in mice to investigate the protective effect of Qingrejiedu Soft Capsules against acute lung injury.Subcutaneous injection of carrageenan was used to establish the tail thrombosis model in mice to investigate the effect of Qingrejiedu Soft Capsules in improving microcirculation.Results:Compared with the model group,the drug groups at 2,4,and 8 g/kg showed prolonged survival time of mice with intravenous injection of LPS at 20 mg/kg and those infected with H1 N1 and decreased mortality rate(P<0.05 orP<0.01).Compared with the normal group,the model group showed increased lung index and lung histopathological score,decreased GSH content in lung tissues,and increased MDA content(P<0.01).After drug administration seven times,the above indexes were improved in the drug groups,especially in the drug group at 8 g/kg(P<0.05 or P<0.01).Compared with the normal group,the model group displayed increased tail thrombosis induced by subcutaneous injection of carrageenan(P<0.01),shortened thrombin time,and increased fibrinogen content(P<0.05 orP<0.01).After drug administration five times,the above indexes were significantly improved in the drug groups at 1,2,and 4 g/kg(P<0.05 orP<0.01).Conclusion:Qingrejiedu Soft Capsules can effectively prolong the survival time of mice infected with influenza virus and those undergoing intravenous injection of high-dose LPS and reduce the mortality.The protective effect against acute lung injury may be achieved by reducing pulmonary oxidative stress,thrombosis,and pulmonary edema.
