艾灸对肝郁脾虚型抑郁大鼠海马体CREB、P-CREB表达影响

摘　　要：目的通过建立肝郁脾虚抑郁症大鼠模型的方法来研究艾灸对肝郁脾虚抑郁症的治疗作用情况以及机理。方法60只大鼠分成6组,每组10只,分别为阳性对照组、阴性对照组、艾灸介入组、氟西汀介入组、艾灸治疗组以及氟西汀治疗组。采用慢性不可预知温和应激方法(CUMS)对实验大鼠进行造模,从造模的第9~18天,艾灸介入组、氟西汀介入组在造模前1 h给予造模期间的介入治疗,在19~30 d分别予氟西汀治疗组氟西汀溶液10 mL/kg以及艾灸治疗组予艾灸百会、肝俞、足三里、太冲等穴位治疗。阳性对照组、阴性对照组、氟西汀介入组和艾灸介入组这4组在第19天造模期间的介入治疗结束之后会对这几组进行处死并且去除这4组的海马组织,氟西汀治疗组和艾灸治疗组在第30天取材,RT-PCR法测环磷腺苷效应元件结合蛋白(CREB)mRNA的含量,Western Blot法检测CREB和p-CREB蛋白含量。结果在阳性对照组大鼠的CREB蛋白表达情况与阴性对照组之间比较之后,前者组内的大鼠CREB蛋白表达明显降低(P<0.05);与阳性对照组相比,艾灸介入组、氟西汀介入组、艾灸治疗组和氟西汀治疗组CREB和p-CREB的含量明显上调(P<0.05)。结论艾灸治疗可通过上调CREB和p-CREB的含量,从而对抑郁症起到预防及治疗作用。Objective To investigate the therapeutic effect and mechanism of moxibustion on hepatic depression and splenic insufficiency depression by creating a rat model of hepatic depression and splenic insufficiency depression.Methods 60 rats were divided into 6 groups with 10 rats in each group:positive control group,negative control group,moxibustion intervention group,fluoxetine intervention group,moxibustion treatment group and fluoxetine treatment group.The experimental rat model was established by CUMS method.From 9 d to 18 d,the moxibustion intervention group and fluoxetine intervention group were given intervention treatment during the modeling period one hour before modeling,and the fluoxetine treatment group was given fluoxetine solution 10 mL/kg and the moxibustion treatment group was given moxibustion Baihui(DU20),Ganshu(BL18),Zusanli(ST36),Taichong(LR3)and other acupoints respectively from 19 d to 30 d.The positive control group,negative control group,fluoxetine intervention group and moxibustion intervention group will be killed and the hippocampal tissue of the four groups will be removed after the intervention treatment on the 19th day.The materials of fluoxetine treatment group and moxibustion treatment group will be taken on the 30th day.The content of CREB mRNA will be measured by RT-PCR,and the content of CREB and p-CREB protein will be detected by Western blot.Results the expression of CREB protein in the positive control group was significantly lower than that in the negative control group(P<0.05);Compared with the positive control group,CREB and p-CREB contents were significantly increased in the moxibustion intervention group,fluoxetine intervention group,moxibustion treatment group,and fluoxetine treatment group(P<0.05).Conclusion Moxibustion can prevent and treat depression by up regulating the contents of CREB and p-CREB.
