摘  要:目的:以EGCs为切入点,研究电针足三里穴对DGP大鼠EGCs的影响,并探讨电针足三里治疗DGP的可能作用机制,为临床治疗DGP提供客观参考依据。方法:将40只健康雄性SD大鼠随机分为造模组(n=32)、正常对照组(n=8),采用腹腔注射链脲佐菌素制备DGP模型,模型成功建立后再将造模组大鼠随机分为DGP模型组和足三里组,每组15只;足三里组针刺双侧足三里并连接电针,DGP模型组与正常对照组给予相同固定方法即可。每周测量大鼠体质量、血糖,治疗结束后检测各组大鼠胃排空率、小肠推进率,q-RTPCR检测S100β、GDNF、MEK1、ERK1mRNA表达水平。结果:与正常对照组比较,DGP模型组大鼠体质量减轻,空腹血糖升高,胃排空率及小肠推进率下降(P<0.05);与DGP模型组比较,足三里组大鼠体质量增加,空腹血糖降低,胃排空率及小肠推进率增加(P<0.05);与正常对照组比较,DGP模型组S100β、GDNF、MEK1、ERK1蛋白表达减少(P<0.05),与DGP模型组比较,足三里组S100β、GDNF、MEK1、ERK1蛋白表达增加(P<0.05)。结论:电针足三里穴可增加DGP大鼠体质量,降低血糖,促进胃排空及小肠推进,其作用机制可能是通过调控ERK信号通路来促进EGCs的增殖分化,从而改善DGP大鼠胃肠功能。Objective:This experiment took EGCs as the starting point to study the effect of electroacupuncture at Zusanli on EGCs in DGP rats,and to explore the possible mechanism of electroacupuncture at Zusanli in the treatment of DGP,so as to provide an objective reference for clinical treatment of DGP.Methods:40healthy male SD rats were randomly divided into the model group(32rats)and the normal control group(8rats).The DGP model was prepared by intraperitoneal injection of streptozotocin.After the model was successfully established,the model rats were randomly divided into the DGP model group and the Zusanli group,with 15rats in each group.The Zusanli group was treated with acupuncture at both sides of Zusanli and connected with electro-acupuncture.The DGP model group and the normal control group were only given the same fixation method.The weight and fasting blood glucose of the rats were measured every week.After the treatment,the materials were collected to detect the gastric emptying rate and the small intestine propulsion rate.S100β,GDNF,MEK1,ERK1 mRNA were detected by q-RTPCR.Results:Compared with the normal control group,The weight of the rats in the DGP group decreased,the fasting blood glucose increased,the gastric emptying rate and the small intestinal propulsion rate decreased(P<0.05).Compared with the DGP group,the weight of the rats in the Zusanli group increased,the fasting blood glucose decreased,the gastric emptying rate and the small intestinal propulsion rate increased(P<0.05).Compared with the normal control group,the expression of S100β,GDNF,MEK1,and ERK1proteins in the DGP model group decreased(P<0.05).Compared with the DGP model group,the expression of S100β,GDNF,MEK1,and ERK1proteins in the Zusanli group increased(P<0.05).Conclusion:The following conclusions can be drawn:Electroacupuncture at Zusanli can increase the DGP rats weight,reduce fasting blood glucose,and promote gastric emptying and small intestine propulsion.Its mechanism may be to promote the proliferation and differentiation of E
