摘 要:目的:研究中药野木瓜镇痛作用的有效部位及其作用机制。方法:采用完全弗氏佐剂诱发的大鼠炎性痛模型,观察野木瓜中的三萜皂苷对大鼠痛阈的影响;并提取出完全弗氏佐剂诱发的炎性痛大鼠的背根神经节,应用Western Blot技术,观察野木瓜三萜皂苷对炎性痛大鼠背根神经节辣椒素受体表达量的影响;最后,应用膜片钳技术,观察野木瓜三萜皂苷对大鼠背根神经节细胞辣椒素诱发的受体电流峰值的抑制效应。结果:野木瓜三萜皂苷在连续给药3天后,能够显著增加完全弗氏佐剂诱发的炎性痛大鼠痛阈,并下调炎性痛大鼠背根神经节辣椒素受体表达量。野木瓜三萜皂苷还能抑制大鼠背根神经节上辣椒素诱发的受体电流峰值,呈浓度依赖关系。结论:三萜皂苷可能是野木瓜镇痛的有效部位之一,其外周镇痛的机制可能与其对辣椒素受体的调控有关。Objective:To study the analgesic effective parts and mechanism of Stauntonia chinensis.Methods:The effect of triterpenoid saponins from Stauntonia chinensis on pain threshold in rats was observed by using complete Freund\s adjuvant induced inflammatory pain model.And the effects of triterpenoid saponins fromStauntonia chinensis on the expression of capsaicin receptor in dorsal root ganglion of rats with inflammatory pain induced by complete Freund\s adjuvant were also observed by Western Blot.Finally,patch clamp technique was used to observe the inhibitory effect of triterpenoid saponins fromStauntonia chinensis on capsaicin induced receptor current peak in rat dorsal root ganglion neurons.Results:The results showed that triterpenoid saponins fromStauntonia chinensis could significantly increase the pain threshold and down regulate the expression of capsaicin receptor in dorsal root ganglion of rats with complete Freund\s adjuvant induced inflammatory pain after 3days of continuous administration.Triterpenoid saponins fromStauntonia chinensis also inhibited capsaicin induced receptor current peak in rat dorsal root ganglion neurons in a concentration dependent manner.Conclusion:These results suggested that triterpenoid saponins should be one of the effective analgesic components of Stauntonia chinensis,and their mechanism of peripheral analgesia may be related to the regulation of capsaicin receptor.
