不同处方祛风活络喷雾剂对佐剂性关节炎大鼠作用研究

摘　　要：目的:探究不同蛇类处方的祛风活络喷雾剂对佐剂性关节炎(AA)模型大鼠的足爪肿胀度和相关炎症因子的影响。方法:SD大鼠随机分为7组,每组8只。参照完全弗氏佐剂造模方法造模,造模后第5天起给药,每隔9d进行关节炎性评分,每隔2d测量致炎侧足爪肿胀度,末次给药3d后取材。采用ELISA法检测血清中白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)和肿瘤坏死因子(TNF-α)的含量。结果:第12~15天,大鼠足趾出现继发性肿大并加重,关节炎性评分超过4分,造模成功。实验结果显示阳性组和5种蛇类祛风活络喷雾剂、乌梢蛇祛风活络喷雾剂、植物药祛风活络喷雾剂分别在给药第15天、第18天、第27天,足爪肿胀度与模型组比较,差异有统计学意义(P<0.05)。与模型组相比,阳性组和5种蛇类制剂能显著抑制IL-1β、IL-6和TNF-α的分泌(P<0.05),乌梢蛇类制剂能显著抑制IL-1β、IL-6的分泌(P<0.05),而植物药仅对IL-1β有显著抑制作用(P<0.05)。结论:5种蛇类处方祛风活络喷雾剂对佐剂性关节炎的炎症损伤起到明显治疗作用,可能与蛇类药材尤其是毒性蛇类药材同时抑制多种炎症因子水平有关。Objective:To investigate the effects of Qufeng Huoluo sprays formulated with different snakes on the paw swelling and inflammatory factors in adjuvant arthritis(AA)model rats.Methods:SD rats were randomly divided into7groups with 8rats in each group.The model was established according to the complete Freund’s adjuvant modeling method.The drug was administered on the 5th day after modeling.The arthritis score was performed every 9days,and the swelling degree of the inflamed paw was measured every 2days,and the materials were collected 3days after the last administration.The interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor(TNF-α)in serum were detected by ELISA.Results:On the 12th to 15th day,the rat’s toes showed secondary swelling and aggravation,and the arthritis score exceeded 4points,indicating that the modeling was successfully established.The results showed that there were significant differences in the paw swelling in the positive group and the 5species of snakes group,the black snake group,and the botanical drug group on the 15th,18th,and 27th days compared with the model group(P<0.05).Compared with the model group,the positive group and five kinds of snake preparations could significantly inhibit the secretion of IL-1β,IL-6and TNF-α(P<0.05).The black snake preparation group could regulate the secretion of IL-1βand IL-6(P<0.05)while the botanical group only decreased the content of IL-1β(P<0.05).Conclusion:The Qufeng Huoluo sprays prescribed for 5kinds of snakes have therapeutic effects on adjuvant arthritis,which may be related to the addition of snake medicinal materials,especially venomous snake medicinal materials,which simultaneously inhibit the levels of various inflammatory factors.
