摘  要:目的观察参竹降糖方对气阴两虚型高血糖大鼠免疫内分泌及骨骼肌胰岛素抵抗的影响。方法根据基础血糖值将SD大鼠分为空白组、复合模型组、阳性药组,以及参竹降糖方高、中、低剂量组共6组,每组15只。采用力竭游泳、甲状腺素(thyroxine,T4)皮下注射、连续高脂饲料合并链脲佐菌素(streptozotoci,STZ)注射复合因素造模。造模同时灌胃给药33天,阳性药组予十味消渴胶囊1.32 g/kg,参竹降糖方高、中、低剂量组分别予参竹降糖方混悬液7.5 g/kg、2.5 g/kg、1.25 g/kg,空白组不做处理,模型对照组给予同体积溶剂。采用比色法测定血清肝糖原、肌糖原、游离脂肪酸(free fatty acid,FFA)、免疫球蛋白G(immunoglobulin G,IgG)、免疫球蛋白M(immunoglobulin M,IgM)、三磷酸腺苷(adenosine triphosphate,ATP)、三磷酸腺苷酶(adenosine triphosphatase,ATPase)免疫及代谢相关指标水平;ELISA法检测促甲状腺激素(thyroid stimulating hormone,TSH)、睾酮(testosterone,T)、雌二醇(estradiol,E2)激素水平及骨骼肌白细胞介素6(interleukin-6,IL-6)、白细胞介素1β(interleukin-1β,IL-1β)、肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)炎症因子含量;比色法检测骨骼肌丙二醛(malondialdehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-PX)氧化应激水平;Western Blot检测骨骼肌胰岛素受体(insulin receptor,InsR)、胰岛素受体底物1(insulin receptor substrate1,IRS1)、磷脂酰肌醇3激酶(phosphatidylinositol 3 kinase,PI3K)、葡萄糖转运蛋白4(glucose transporter 4,GLUT4)蛋白表达水平。结果与模型组相比,参竹降糖方各剂量组大鼠血清FFA、E2、TSH,骨骼肌MDA、IL-6、IL-1β、TNF-α水平显著降低(P<0.01),血清肝糖原、肌糖原、IgG、IgM、T、ATP、ATP酶和骨骼肌SOD、GSH-PX,以及InsR、IRS1、PI3K、GLUT4蛋白表达水平显著升高(P<0.05,P<0.01)。结论参竹降糖方能够改善气阴两虚�Objective To observe the effects of Shenzhu Jiangtang recipe on oxidative stress-inflammatory factors-insulin resistance in hyperglycemic rats with Qi and Yin deficiency.Methods According to the basal blood glucose value,SD rats were divided into 6 groups:the blank group,the compound model group,the positive drug group,and the Shenzhu Jiangtang recipe high-dose,medium-dose and low-dose groups,with 15 rats in each group.The model was constructed by exhausting swimming,subcutaneous injection of thyroxine(T4),continuous high-fat diet combined with streptozotocin(STZ)injection.At the same time,the drug was given by gavage for 33 days,the rats in the positive drug group was given Shiwei Xiaoke capsule 1.32 g/kg,the rats in the Shenzhu Jiangtang recipe high,medium and low dose groups were given Shenzhu Jiangtang recipe suspension 7.5 g/kg,2.5 g/kg and 1.25 g/kg respectively,while the rats in the blank group was not treated,and the rats in the model control group was given the same volume of solvent.Colorimetric method was used to determine indicators related to metabolism level and immune function,such as serum liver glycogen,muscle glycogen,free fatty acid(FFA),immunoglobulin G(IgG),immunoglobulin M(IgM),adenosine triphosphate(ATP),adenosine triphosphatase(ATPase)indicators.The levels of thyroid stimulating hormone(TSH),testosterone(T),estradiol(E2),interleukin-6(IL-6),interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)in skeletal muscle were detected by ELISA.The expression levels of skeletal muscle malondialdehyde(MDA),superoxide dismutase(SOD)and glutathione peroxidase(GSH-PX)were detected by colorimetric method.Skeletal muscle insulin receptor(InsR),insulin receptor substrate 1(IRS1),phosphatidylinositol 3 kinase(PI3K),glucose transporter 4(GLUT4)were detected by Western Blot.Results Compared with model group,serum FFA,E2,TSH,skeletal muscle MDA,IL6,IL1βand TNFαlevels in Shenzhu Jiangtang recipe groups were significantly decreased(P<0.01).Serum liver glycogen,muscle glycogen,IgG,IgM,T,ATP,ATPase,skeletal
