摘  要:目的通过体外实验探讨黄芪多糖(astragalus polysaccharides,APS)与丹酚酸B(salvianolic acid B,Sal B)配伍对转化生长因子-β1(transforming growth factor-β1,TGF-β1)诱导的肺上皮细胞间质转化的影响,并探讨黄芪与丹参组分配伍抗肺纤维化作用的可能机制。方法设空白组、黄芪多糖组(60 mg/L)、丹酚酸B组(60 mg/L)、配伍组(APS 60 mg/L+Sal B 60 mg/L),利用MTS方法检测对TGF-β1诱导的人肺泡上皮细胞(A549)增殖的影响;采用实时荧光定量PCR法和蛋白免疫印迹法(western blot)检测它们对TGF-β1诱导A549细胞的E-钙黏蛋白(E-Cadherin,E-Cad)、α-平滑肌肌动蛋白(α-Smooth muscle actin,α-SMA)和波形蛋白(vimentin,Vim)表达影响;并采用酶联免疫吸附法(ELISA)检测药物对TGF-β1诱导A549细胞的纤连蛋白(fibronectin,FN)和I型胶原蛋白(Collagen-I,Col-I)表达的影响。结果黄芪多糖组、丹酚酸B组和配伍组对A549细胞的增殖均无影响,但丹酚酸B组、配伍组对TGF-β1诱导A549细胞增殖有抑制作用;Western blot结果显示:配伍组E-Cad的蛋白表达显著增加(P<0.01),对α-SMA和Vim的表达具有抑制作用(P<0.05);ELISA结果示配伍组对TGF-β1诱导A549细胞FN和Col-I的表达具有抑制作用。结论黄芪多糖与丹酚酸B配伍对TGF-β1诱导的肺上皮细胞间质转化的具有抑制作用,该配伍组合具有治疗肺纤维化的应用前景。Objective The effects of astragalus polysaccharide and salvianolic acid B on TGF-β1 induced pulmonary epithelial mesenchymal transition were investigated in vitro,and the possible mechanism of Astragalus membranaceus and salvianolic acid B on pulmonary fibrosis was discussed.Methods The blank group,the astragalus polysaccharide group,the salvianolic acid B group and the astragalus polysaccharide and the salvianolic acid B compatibility group were set up in the experiment.The effects of them on the proliferation of A549 cells induced by TGF-β1 were detected by MTS method.The methods of Realtime PCR and Western blot were used to detect the expressions of E-cad,α-SMA and Vim in A549 cells induced by TGF-β1.Finally,the method of ELISA was used to detect the effects of drugs on the expression of FN and Col-I in A549 cells induced by TGF-β1.Results The astragalus polysaccharide group,the salvianolic acid B group and the compatibility group had no effect on the proliferation of A549 cells,but the salvianolic acid B group and the astragalus polysaccharide plus salvianolic acid B group inhibited the proliferation of A549 cells induced by TGF-β1.The resutls of Western blot results showed that the protein expression of E-cad was significantly increased(P<0.01),and the expression ofα-SMA and Vim was inhibited(P<0.01).The results of ELISA showed that the combination group could inhibit the expression of FN and Col-I in A549 cells induced by TGF-β1.Conclusion The combination of astragalus polysaccharide and salvianolic acid B has inhibitory effect on TGF-β1 induced pulmonary epithelial mesenchymal transition.The combination of astragalus polysaccharide and salvianolic acid B has the application prospect in the treatment of pulmonary fibrosis.
