苓甘五味姜辛汤对慢性支气管炎的作用机制

摘　　要：目的研究苓甘五味姜辛汤治疗寒饮蕴肺型慢支炎大鼠模型的生物学机制。方法将雄性SD大鼠随机分组,每组10只,分别为空白组、慢支炎组、寒饮蕴肺组与中药治疗组。除空白组外,各组均气管注入200μg(1μg/μl)脂多糖并予以烟熏,寒饮蕴肺组与中药干预组烟熏后予以寒冷刺激,持续30 d。自第16天起,空白组和慢支炎组予以生理盐水灌胃,中药干预组予苓甘五味姜辛汤1 g/ml灌服,持续2周,检测血清中IL-2、IL-4与肺、支气管中AQP-1的表达水平。结果IL-2、IL-4含量,慢支炎与寒饮蕴肺组比空白组高(P<0.05),中药组比寒饮蕴肺组含量低(P<0.05);AQP-1的表达水平,慢支炎组与寒饮蕴肺组比空白组高(P<0.05),中药组比寒饮蕴肺组含量高。结论苓甘五味姜辛汤可通过降低IL-2、IL-4含量和提高AQP-1的表达,起到平衡调节炎症因子和气道液体的作用,达到治疗慢性支气管炎的目的。Objective To investigate the biological mechanism of Linggan Wuwei Jiangxin decoction in the treatment of rat model of cold-drinking lung-embedded chronic bronchitis.Methods Male SD rats were randomly grouped into 10 groups,including blank group,laryn gitis group,cold-drinking lung group and herbal treatment group.All groups except the blank group were injected with 200 ug(1μg/μl)of lipopolysaccharide and fumigated,while the cold-drinking lung group and the herbal intervention group were fumigated and then cold-stimulated for 30 days.From day 16 onwards,the blank group and the chronic bronchitis group were given saline gavage,while the Chinese herbal medicine intervention group was given Linggan Wuwei Jiangxin decoction 1 g/ml for a fortnight,and the expression levels of IL-2 and IL-4 in serum and AOP-1 in lung and bronchus were measured.Results The levels of IL-2 and IL-4 were higher in the group with chronic bronchitis and cold-drinking lung than in the blank group(P<0.05),and lower in the group with traditional Chinese medicine than in the group with cold-drinking lung(P<0.05);the expression level of AOP-1 was higher in the group with chronic bronchitis and cold-drinking lung than in the blank group(P<0.05),and higher in the group with traditional Chinese medicine than in the group with cold-drinking lung.Conclusion Linggan Wuwei Jiangxin decoction can play a role in balancing and regulating inflammatory factors and airway fluid by reducing IL-2 and IL-4 levels and increasing the expression of AOP-1 to achieve the purpose of treating chronic bronchitis.
