摘 要:目的探讨趋化素样因子超家族成员1(CKLFSF1)基因表达与冠心病发生风险的相关性及相关机制。方法从2020年5月至2021年7月间在河南大学附属南阳市中心医院行冠状动脉造影检查的患者中选取1支及以上血管直径狭窄程度>50%的患者175例为冠心病组;未达到上述诊断标准的非冠心病患者140例为对照组。收集患者的人口学特征及合并症情况,采集空腹外周血,检测血脂水平、血浆CKLFSF1蛋白表达水平;RT-PCR检测外周血CKLFSF1mRNA表达水平。常规培养人主动脉内皮细胞(HAECs)并进行分组:正常HAECs组(常规培养,不做质粒转染处理);过表达CKLFSF1HAECs组(转染过表达CKLFSF1腺病毒质粒);低表达CKLFSF1HAECs组(转染低表达CKLFSF1腺病毒质粒)。同时常规培养人单核细胞THP-1待用。进行细胞Transwells迁移实验和黏附实验评价CKLFSF1基因对THP-1趋化能力的影响。Western blot检测各组HAECs中VCAM-1和E-选择素蛋白表达水平。结果冠心病组患者CKLFSF1mRNA和血浆蛋白表达水平明显高于对照组,差异有统计学意义(P<0.01)。年龄、吸烟史、糖尿病、高血压、CKLFSF1是冠心病发生的独立影响因素。过表达CKLFSF1HAECs组中THP-1细胞迁移数量明显高于正常HAECs组,低表达CKLFSF1 HAECs组中THP-1细胞迁移数量明显低于正常HAECs组,差异均有统计学意义(P<0.01)。过表达CKLFSF1 HAECs组中THP-1细胞黏附数量明显高于正常HAECs组,低表达CKLFSF1HAECs组中THP-1细胞黏附数量明显低于正常HAECs组,差异均有统计学意义(P<0.01)。过表达CKLFSF1 HAECs组中VCAM-1、E-选择素蛋白表达水平明显高于正常HAECs组,低表达CKLFSF1HAECs组中VCAM-1、E-选择素蛋白表达水平明显低于正常HAECs组,差异均有统计学意义(P<0.01)。结论CKLFSF1基因高表达与冠心病的发生密切相关;CKLFSF1基因高表达能够通过上调VCAM-1、E-选择素表达来促进人单核细胞迁移和黏附,诱导人主动脉内皮细胞增殖,�Objective To investigate the correlation and mechanism between the gene expression of chemokine like factor superfamily member 1(CKLFSF1)and the risk of coronary heart disease.Methods From May 2020to July2021,175patients with diameter stenosis of one or more vessels>50%were selected as the coronary heart disease group;140non coronary heart disease patients who did not meet the above diagnostic criteria were the control group.The demographic characteristics and complications of patients were collected.Fasting peripheral blood was collected to detect the level of blood lipid and the expression of plasma CKLFSF1protein;The expression of CKLFSF1mRNA in peripheral blood was detected by RT-PCR.Human aortic endothelial cells(HAECs)were routinely cultured and divided into normal HAECs group(routine culture without plasmid transfection);Overexpression of CKLFSF1HAECs group(transfection of adenovirus plasmid overexpression of CKLFSF1);Low expression of CKLFSF1HAECs group(transfected with low expression of CKLFSF1adenovirus plasmid).Meanwhile,human monocytes(THP-1)were routinely cultured for use.Cell transwells Migration Experiment and adhesion experiment were carried out to evaluate the effect of CKLFSF1gene on THP-1chemotaxis.The expression levels of VCAM-1and E-selectin protein in HAECs were detected by Western blot.Results The expression levels of CKLFSF1mRNA and plasma protein in patients with coronary heart disease were significantly higher than those in the control group(P<0.01).Age,smoking history,diabetes,hypertension and CKLFSF1were independent factors affecting CHD.The migration number of THP-1cells in over expressing CKLFSF1HAECs group was significantly higher than that in normal HAECs group,and the migration number of THP-1cells in low expressing CKLFSF1 HAECs group was significantly lower than that in normal HAECs group(P<0.01).The number of THP-1Cell adhesion in over expressing CKLFSF1 HAECs group was significantly higher than that in normal HAECs group,and the number of THP-1Cell adhesion in low expressing C
