黄芩苷元对妊娠期糖尿病大鼠妊娠结局及免疫调节的实验研究

摘　　要：目的探讨黄芩苷元对妊娠期糖尿病(GDM)大鼠妊娠结局的影响。方法SD雌性大鼠采用高脂饮食联合STZ建立妊娠期糖尿病大鼠模型,模型加药组大鼠口服黄芩苷元治疗。计算各组大鼠子宫指数、卵巢指数、胚胎总数和流产率,HE染色观察肝组织病理损伤。血糖仪检测空腹血糖水平,全自动生化分析仪检测谷丙转氨酶(ALT)、谷草转氨酶(AST)、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)水平,试剂盒检测超氧化物歧化酶(SOD)、乳酸脱氢酶(LDH)和丙二醛(MDA)含量水平,ELISA检测外周血和肝组织中肿瘤坏死因子α(TNF-α)、白介素-10(IL-10)含量,Western blot检测Bax/Bcl-2、TNF-α和IL-10蛋白水平。结果与模型组大鼠比较,50、100 mg/kg黄芩苷元组和二甲双胍200 mg/kg组大鼠子宫指数、卵巢指数、胚胎总数显著增多(P<0.05),流产率显著降低(P<0.05),肝组织病理损伤减轻,空腹血糖水平显著降低(P<0.05),ALT、AST、TC、TG和LDL含量显著降低(P<0.05),HDL含量显著升高(P<0.05),SOD含量升高,MDA和LDH含量降低(P<0.05),Bax/Bcl-2和TNF-α含量水平显著下调(P<0.05),IL-10含量水平显著上调(P<0.05)。结论黄芩苷元对妊娠期糖尿病大鼠妊娠结局具有调控作用。Objective To investigate the effect of baicalein on pregnancy outcome in gestational diabetes(GDM)rats.Methods SD female rats were treated with high-fat diet combined with STZ to establish gestational diabetes rat models.The rats in the model plus drug group were treated with baicalein orally.Calculate the uterine index,ovarian index,total number of embryos and abortion rate of rats in each group,and observe the pathological damage of liver tissue by HE staining.Fasting blood glucose level was detected by blood glucose analyzer,automatic biochemical analyzer to detect alanine aminotransferase(ALT),aspartate aminotransferase(AST),total cholesterol(TC),triglycerides(TG),high density lipoprotein(HDL),low density lipoprotein(LDL)levels,reagents The box detects the levels of superoxide dismutase(SOD),lactate dehydrogenase(LDH)and malondialdehyde(MDA),and ELISA detects tumor necrosis factor-α(TNF-α)and interleukin-10 in peripheral blood and liver tissue.IL-10)content,Western blot was used to detect Bax/Bcl-2,TNF-αand IL-10 protein levels.Results Compared with model group,uterine index,ovarian index and total number of embryos in 50 and 100 mg/kg Baicalein groups and 200 mg/kg metformin groups were significantly increased(P<0.05),abortion rate was significantly decreased(P<0.05),pathological damage of liver tissue was alleviated,and fasting blood glucose level was significantly decreased(P<0.05).The contents of ALT,AST,TC,TG and LDL were significantly decreased(P<0.05),HDL content was significantly increased(P<0.05),SOD content was increased,MDA and LDH contents were decreased(P<0.05),Bax/Bcl-2 and TNF-αcontents were significantly decreased(P<0.05).Il-10 content was significantly up-regulated(P<0.05).Conclusion baicalein has a regulatory effect on the pregnancy outcome of gestational diabetic rats.
