摘　　要：目的:研究多梳样蛋白2(PCL2)对胶质瘤血管生成的影响及相关机制。方法:癌症基因组图谱(TCGA)分析PCL2基因在多种恶性肿瘤中的表达;免疫组织化学染色观察PCL2基因在胶质瘤中的表达,并分析PCL2与肿瘤厚壁血管形成的相关性;构建PCL2重组腺病毒,感染U87-MG细胞(MOI=20∶1、50∶1、100∶1),Western blot检测PCL2及zeste基因增强子同源物2(EZH2)、血红素氧合酶1(HO-1)和核因子E2相关因子(Nrf2)表达。结果:TCGA数据库分析及免疫组化结果表明PCL2在多种恶性肿瘤中表达异常,在高、低级别胶质瘤中呈高表达,且与胶质瘤厚壁血管形成呈正相关。PCL2上调了EZH2、Nrf2和HO-1表达。结论:PCL2与胶质瘤血管生成有关,并对PCL2血管生成起正向促进作用。Objective:To study effect of polycomb-like protein 2(PCL2)on angiogenesis in glioma and its mechanism.Methods:Expression of PCL2 gene in many kinds of malignant tumors was analyzed by cancer genome map(TCGA);expression of PCL2 gene in glioma was observed by immunohistochemical staining,and correlation between PCL2 and tumor thick wall angiogenesis was analyzed. PCL2 recombinant adenovirus was constructed and infected U87-MG cells with gradients set at MOI=20∶1,50∶1 and100∶1. Western blot was used to detect expressions of PCL2,enhancer of zeste homolog 2(EZH2),heme oxygenase1(HO-1)and nuclear factor erythroid 2-related factor 2(Nrf2).Results:TCGA database analysis and immunohistochemical results show that expression of PCL2 was abnormal in many kinds of malignant tumors,which was high expression in high and low grade of gliomas,and positively corrected with thick wall vessels. PCL2 increased expressions of EZH2,Nrf2 and HO-1.Conclusion:PCL2 is relates to angiogenesis of glioma and plays a positive role in promoting angiogenesis.
