摘  要:目的:探讨疏风通络方调控STAT1信号通路对支气管哮喘小鼠模型Th1/Th2失衡的影响。方法:实验级小鼠30只,随机分为对照组、模型组、IL-27组、疏风通络方组和疏风通络方+STAT1抑制剂组,每组6只。观察5组小鼠一般情况;记录各组小鼠咳嗽情况;比较血清及灌洗液中IL-4及IFN-γ表达;流式细胞术检测Th1/Th2;HE染色观察小鼠肺组织炎症情况;Western blot检测pSTAT1及STAT1蛋白表达水平。结果:对照组小鼠呼吸平稳、无喘息,精神状况正常,运动良好,毛发颜色正常;模型组呼吸困难伴喘息,精神萎靡,蜷缩不动,毛发暗淡;IL-27组、疏风通络方组造模后同模型组,用药后呼吸逐渐恢复,喘息减少,精神状态好转,开始活动;疏风通络方+STAT1抑制剂组造模后同模型组,用药后未见明显改变。模型组(12.55±3.21)咳嗽次数较对照组(1.31±0.26)增加;IL-27组(5.89±1.27)和疏风通络方组(4.76±1.33)咳嗽次数较模型组减少,疏风通络方+STAT1抑制剂组(9.13±1.25)咳嗽次数较疏风通络方组增加。模型组血清(29.98±6.35)及灌洗液(16.59±4.49)IL-4水平较对照组(10.12±0.91、1.35±0.67)升高,IFN-γ水平降低(12.39±4.31、14.36±3.96 vs 21.35±4.73、32.56±4.51);IL-27组(11.23±2.56、1.15±0.52)和疏风通络方组(12.96±2.58、2.21±1.61)血清及灌洗液IL-4较模型组降低,IFN-γ水平升高(17.09±2.35、20.15±2.29vs 18.63±1.35、26.56±3.39);疏风通络方+STAT1抑制剂组血清(20.56±4.32)及灌洗液(12.68±3.56)IL-4较疏风通络方组升高,IFN-γ水平降低(13.55±2.91、17.84±4.42)。模型组(0.20±0.03)小鼠Th1/Th2较对照组(1.15±0.18)降低;IL-27组(0.62±0.04)和疏风通络方组(0.67±0.02)小鼠Th1/Th2较模型组升高;疏风通络方+STAT1抑制剂组(0.32±0.01)小鼠Th1/Th2较疏风通络方组降低。对照组小鼠肺泡、支气管结构正常;模型组肺泡间隙变大,出现明显炎症反应,支气管上皮细胞脱落;IL-27组和疏风通络方组较模型组炎症反应�Objective:To investigate effect of Shufeng Tongluo prescription regulating STAT1 signaling pathway on Th1/Th2imbalance in mice with bronchial asthma.Methods:Thirty experimental mice were stochastic divided into control group,model group,IL-27 group,Shufeng Tongluo prescription group and Shufeng Tongluo prescription+STAT1 inhibitor group,with 6 mice in each group.Observd status of mice in five groups;tussis of mice in each group was recorded;expressions of IL-4 and IFN-γin serum and bronchoalveolar lavage fluid were compared.Th1/Th2 was detected by flow cytometry.HE staining was used to survey inflammation of lung tissue of mice.Western blot was used to detect pSTAT1 and STAT1 protein expression levels.Results:In control group,breathing was stable without wheezing,the mental condition was normal,the exercise was good,and the fur tinct was regular.Model group had dyspnea accompanied by wheezing,listlessness of spirit,curled up immobility and dark hair.IL-27 group and Shufeng Tongluo prescription group were the same as model group after modeling,after medication,breathing was gradually recovered,wheezing was decreased,mental state was improved and activities began.Shufeng Tongluo prescription+STAT1 inhibitor group was the same as model group after modeling,and no significant changes were surveyed after drug.Cough frequency of model group(12.55±3.21)was higher than that of control group(1.31±0.26).Compared with model group,cough frequency of IL-27 group(5.89±1.27)and Shufeng Tongluo prescription group(4.76±1.33)were decreased,and cough frequency of Shufeng Tongluo prescription+STAT1 inhibitor group(9.13±1.25)was increased compared with Shufeng Tongluo prescription group.Levels of IL-4 in serum of model group(29.98±6.35)and lavage fluid(16.59±4.49)were higher than that of control group(10.12±0.91,1.35±0.67).Level of IFN-γwas decreased(12.39±4.31,14.36±3.96 vs 21.35±4.73,32.56±4.51).Serum and lavage fluid IL-4 in IL-27 group(11.23±2.56,1.15±0.52)and Shufeng Tongluo prescription group(12.96±2.58,2.21±
