摘 要:目的观察蒙药当贡-3对急性心肌梗死(AMI)大鼠心肌组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、丙二醛(MDA)及血清中乳酸脱氢酶(LDH)、肌酸磷酸激酶(CPK)含量的影响。方法100只SPF级别SD雄性大鼠,随机选取80只采用冠脉结扎法建立AMI模型,将建模成功的50只大鼠随机分为模型组、西药组(氟伐他汀)和当贡-3高、中、低剂量组,每组10只,另取10只大鼠作为假手术组,剩余10只留作空白组,各组于术后24 h分别灌胃相应药物,模型组、假手术组和空白组灌胃等体积的蒸馏水,每日1次,连续灌胃4周。实验结束后取材,用生化法检测各组大鼠心肌组织SOD、CAT、GSH-Px、MDA及血清LDH、CPK的含量。结果与假手术组相比,模型组大鼠的血清中LDH、CPK的含量升高,心肌组织中SOD、GSH-Px和CAT活性降低,MDA含量升高(P<0.05)。与模型组比较,当贡-3高、中、低剂量组及氟伐他汀组大鼠的血清中LDH、CPK的含量降低,心肌组织中SOD、GSH-Px和CAT活性增强,MDA含量减少(P<0.05)。结论蒙药当贡-3对急性心肌梗死大鼠心肌具有保护作用,且这种保护作用可能与调节氧化应激反应有关。Objective:To explore the effects of Mongolia medicine Danggong-3 on myocardial tissue SOD,GSH-Px,CAT,MDA,LDH and CPK in acute myocardial infarction(AMI). Methods:A total of 100 SPF male SD rats were raised;80 SD rats were randomly selected to establish AMI model by coronary ligation,and 50 successfully modeled rats were randomly divided into model group,Danggong-3 high-dose,medium-dose and low-dose groups,western medicine group(fluvastatin),with 10 rats in each group;another 10 rats were selected as sham operation group. The remaining 10 were left as blank control group. Each group was given corresponding drugs by intragastric administration 24 hours after operation. The model group,sham operation group and blank control group were given equal volume of distilled water by intragastric administration,once a day,for 4 weeks. At the end of the experiment,the SOD,CAT,GSH-Px,MDA and LDH and CPK contents in myocardium of rats in each group were determined by biochemical method. Results:Compared with sham operation group,the contents of LDH and CPK in serum of model group were increased,the activities of SOD,GSH-Px and CAT in myocardial tissue were decreased,and the content of MDA was increased(P < 0.05). Compared with model group,when Danggong-3 highdose,medium-dose and low-dose groups and fluvastatin group,the contents of LDH and CPK in serum were decreased,the activities of SOD,GSH-Px and CAT in myocardial tissue were increased,and the content of MDA was decreased,the differences of above indexes were statistically significant(P < 0.05). Conclusion:Mongolian medicine Danggong-3 has protective effect on myocardium of acute myocardial infarction rats,and this protective effect is related to the regulation of oxidative stress response.
