冰水潮湿诱发的脾虚对DBA/1小鼠胶原诱导型关节炎的影响及参苓白术散的“以方测证”

摘　　要：目的:探究冰水潮湿诱发的脾虚证对胶原诱导型关节炎(CIA)病情的影响。方法:首先予饮用冰水和潮湿环境[湿度(80±5)%]诱发DBA/1小鼠脾虚证,再采用牛源性Ⅱ型胶原建立CIA模型,后予以参苓白术散颗粒治疗,比较不同处理下各组DBA/1小鼠的脾虚证及关节炎病情差异。结果:DBA/1小鼠饮用冰水和处于潮湿环境30 d后,血清α淀粉酶、胃泌素和胰蛋白酶较空白对照组显著下降(P<0.01,P<0.05),表明小鼠出现脾虚证;与CIA模型组小鼠比较,冰水潮湿组小鼠体内血清IL-6、G-CSF和关节炎症评分(第65天)上调(P<0.05);参苓白术散治疗后,与冰水潮湿组比较,可上调α淀粉酶和胃泌素(P<0.05),缓解关节炎症状及下调血清anti-CⅡ抗体、G-CSF和IL-6水平(P<0.05,P<0.01)。结论:饮用冰水和外部潮湿环境可成功诱发小鼠脾虚证,且脾虚证加重CIA小鼠关节炎病情,参苓白术散可缓解脾虚证CIA小鼠关节炎症状。Objective:To explore the effects of humidity and ice water induced spleen deficiency on CIA disease.Methods:Firstly,ice water and humid environment[(80±5)%]were used to induce spleen deficiency syndrome in DBA/1 mice,then bovine type II collagen was used to induce CIA model and Shenling Baizhu Powder was used to treat the mice.Besides,this study compared the difference in spleen deficiency syndrome and arthritis in DBA/1 mice between groups.Results:After drinking ice water and being exposed to high humidity for 30 days,compared with the blank control group,DBA/1 mice showed spleen deficiency syndrome including decreased levels of serumα-amylase,gastrin,and trypsin(P<0.01,P<0.05).Compared with CIA model group,the ice water and humidity group mice increased serum IL-6,G-CSF,and arthritis scores(on the 65th day)(P<0.05).In addition,compared with the ice water and humidity group,Shenling Baizhu Powder could restore spleen-deficiency syndrome by upregulatingα-amylase and gastrin(P<0.05),alleviate arthritis by downregulating arthritis score,joint swelling,serum antiCII,IL-6,and C-GSF(P<0.05,P<0.01).Conclusion:Drinking ice water and high humidity successfully induces spleen deficiency syndrome in DBA/1 mice,and spleen deficiency syndrome can aggravate arthritis in mice,Shenling Baizhu Powder can alleviate in CIA mice with spleen deficiency syndrome.
