摘  要:目的:研究狼疮定及狼疮定联合激素对系统性红斑狼疮(SLE)小鼠抗菌肽-浆细胞样树突细胞(p DC)-干扰素α(IFNα)环路的影响。方法:28只MRL/lpr狼疮小鼠随机分为模型组、中药组、中药+激素组、激素组,每组7只,以7只MRL/MpJ小鼠为空白组。运用ELISA法检测用药后各组小鼠血清中抗双链DNA(抗ds-DNA)、抗菌肽、IFNα含量;使用RT-qPCR技术检测狼疮小鼠脾脏中性粒细胞中抗菌肽Camp mRNA的表达;采用流式细胞术检测狼疮小鼠脾脏B220^(+)CD11c^(int)CD317^(+)pDC细胞含量;使用免疫荧光染色观察小鼠肾脏抗菌肽表达情况。结果:与空白组比较,模型组小鼠血清抗ds-DNA、抗菌肽、IFNα含量显著增多(P<0.01);与模型组比较,各用药组抗ds-DNA、抗菌肽及IFNα含量均显著降低(除中药组抗菌肽外)(P<0.01,P<0.05)。与空白组比较,模型组Camp mRNA表达量显著增多(P<0.01);与模型组比较,中药组和激素组Camp mRNA表达均显著减少(P<0.01)。与空白组比较,模型组小鼠脾脏中B220+CD11c^(int)CD317^(+)p DC细胞含量显著增多(P<0.01);与模型组比较,各用药组小鼠脾脏中B220^(+)CD11c^(int)CD317^(+)p DC细胞含量显著减少(P<0.01)。空白组小鼠肾脏基本不表达抗菌肽,模型组肾脏肾小球高表达抗菌肽;与模型组比较,中药组、中药+激素组、激素组小鼠抗菌肽表达减少,在肾小球上沉积减少。结论:狼疮定及狼疮定联合激素可能通过对SLE抗菌肽-p DC-IFNα环路产生影响,从而有益于SLE病情改善。Objective:To investigate the effects of Langchuangding(LCD)and LCD combined with glucocorticoid(GC)on Camp-pDC-IFNαloop in systemic lupus erythematosus(SLE)mice.Methods:Twenty-eight MRL/lpr mice were randomly divided into four groups:model group,LCD group,LCD+GC group and GC group,7 mice in each group.Seven MRL/MpJ mice were used as blank group.ELISA was used to measure anti-ds-DNA,Cathelicidin and IFNαin serum of mice,RT-qPCR was used to detect the expression of Camp mRNA in spleen neutrophils,flow cytometry was used to detect the content of B220^(+)CD11c^(int)CD317^(+)pDC cells in spleen of SLE mice,and immunofluorescence was used to observe the expression of Cathelicidin in kidney of mice.Results:Compared with blank group,anti-ds-DNA,Cathelicidin and IFNαin serum of model group were significantly increased(P<0.01).Compared with model group,anti-ds-DNA,Cathelicidin and IFNαin serum of three treated groups(except Cathelicidin level in the LCD group)were significantly decreased(P<0.01,P<0.05).Compared with blank group,Camp mRNA in spleen neutrophils in model group was increased(P<0.01).Compared with model group,Camp mRNA in spleen neutrophils in the LCD group and GC group decreased(P<0.01).Compared with blank group,the content of B220^(+)CD11c^(int)CD317^(+)pDC in spleen of model group was statistically increased(P<0.01).Compared with model group,the content of B220^(+)CD11c^(int)CD317^(+)pDC in spleen of mice in three treated groups were observably decreased(P<0.01).Blank group was no expression of Cathelicidin in kidney,model group was high expression of Cathelicidin in kidney.Compared with the model group,the expression of Cathelicidin and the deposition of Cathelicidin on glomeruli were decreased in LCD group,LCD+GC group and GC group.Conclusion:LCD and LCD combined with GC may contribute to prevent the progression of SLE by influencing the lupus mice Camp-pDC-IFNαloop.
