益肾祛浊方对PCOS-IR模型大鼠IGF-1表达的影响

摘　　要：目的:探索益肾祛浊方对多囊卵巢综合征胰岛素抵抗(PCOS-IR)模型大鼠垂体及卵巢组织中胰岛素样生长因子1(IGF-1)受体蛋白及IGF-1 mRNA表达的影响。方法:将84只SD雌性大鼠随机分为7组:空白组、模型组、单纯益肾组、益肾祛浊方组、二甲双胍组、达英-35组、达英-35联合二甲双胍组,每组12只。采用来曲唑灌胃联合高脂饲料喂养法构建PCOS-IR大鼠模型。除空白组外,其余各组造模后予相应药物灌胃2周,治疗结束后取大鼠垂体、卵巢组织。免疫荧光法及Western Blot法检测各组大鼠垂体、卵巢组织中IGF-1R蛋白表达差异;荧光定量PCR法检测大鼠垂体、卵巢组织中IGF-1 mRNA表达差异。结果:模型组大鼠垂体、卵巢组织中IGF-1R蛋白及IGF-1 mRNA的表达较空白组显著降低(P<0.05,P<0.01);益肾祛浊组大鼠垂体和卵巢组织IGF-1R蛋白及IGF-1mRNA的表达较模型组显著升高(P<0.05,P<0.01),整体效果优于单纯益肾组及二甲双胍组。结论:益肾祛浊方治疗PCOS-IR的作用机制可能与上调IGF-1R蛋白及其mRNA在垂体和卵巢上的表达密切相关。Objective:To explore the effect of Yishen Quzhuo Recipe on polycystic ovary syndrome-insulin resistance(PCOS-IR)model rats,to explore effects of insulin-like growth factor 1(IGF-1)receptor protein and IGF-1 mRNA expression in pituitary and ovarian tissues.Methods:A total of 84 SD female rats were randomly divided into 7 groups:blank group,model group,simple Yishen group,Yishen Quzhuo Recipe group,metformin group,Diane-35 group,Diane-35 combined with metformin group,12 per group.PCOS-IR model was established by intragastric administration of letrozole combined with high-fat diet.Except for the blank group,the other groups were given the corresponding drugs by gavage for 2 weeks after modeling,and the pituitary and ovarian tissues of the rats were collected after the treatment.Immunofluorescence method and Western Blot method were used to detect the difference of IGF-1 receptor protein expression in pituitary gland and ovarian tissue of rats in each group;fluorescence quantitative PCR method was used to detect the difference of IGF-1 mRNA expression in rat pituitary gland and ovarian tissue.Results:The expression of IGF-1 receptor protein and IGF-1 mRNA in the pituitary gland and ovary of rats in the model group was significantly lower than that in the blank group(P<0.05,P<0.01).The expression of protein and IGF-1 mRNA was higher than that of the model group(P<0.05,P<0.01),and the overall effect was better than that of the simple Yishen group and the metformin group.Conclusion:The mechanism of Yishen Quzhuo Recipe in the treatment of PCOS-IR may be closely related to the up-regulation of the expression of IGF-1 receptor protein and its mRNA in pituitary and ovary.
