摘  要:目的:观察蔡氏内异Ⅲ方对人子宫内膜异位症(EMS)内膜间质细胞中miR-141-3p表达的影响,探讨蔡氏内异Ⅲ方治疗EMS的作用机制。方法:原代分离培养EMS患者在位、异位以及正常人子宫内膜间质细胞,分为在位内膜组(Eu组)和异位内膜组(Ec组),每组分别设模型组(Model组)、西药组(DNG组)(50 nmol/L地诺孕素)和中药组(TCM组)(4 mg/L蔡式内异Ⅲ方药液),另外设置正常对照组。各组加入相应药物干预后,采用RT-PCR及Western Blot检测各组中miR-141-3p及TGF-β1、Smad2蛋白的表达,流式细胞仪检测细胞的凋亡情况。结果:与Model组比较,Eu组和Ec组中miR-141-3p表达水平显著升高(P<0.01),TGF-β1和Smad2蛋白表达水平降低(P<0.05),同时各给药组均能够显著促进细胞凋亡(P<0.01)。结论:蔡氏内异Ⅲ方通过提高EMS内膜间质细胞中miR-141-3p表达水平,抑制下游TGF-β1/Smad2信号通路的活性,促进细胞凋亡,发挥治疗作用。Objective:To observe the effect of CAIs’NeiyiⅢPrescription on the expression of miR-141-3p in endometrial stromal cells of human endometriosis(EMS),and to explore the mechanism of CAIs’NeiyiⅡPrescription in the treatment of EMS.Methods:Culturing eutopic,ectopic endometrial stromal cells isolated from endometriosis patients and normal human EMS stromal cells.The above cells were divided into Eu group and Ec group,each group was set with model group(Model group),western medicine group(DNG group)(50 nmol/L)and CAIs’NeiyiⅡPrescription group(TCM group)(4 mg/L),and also set normal control group.After that,the expressions of miR-141-3p,TGF-β1 and Smad2 proteins in each group were detected by RT-PCR and Western Blot,and cell apoptosis was detected by flow cytometry.Results:Compared with the Model group,in the Eu group and Ec group,the expression level of miR-141-3p was significantly increased(P<0.01),and the protein contents of TGF-β1 and Smad2 were decreased(P<0.05).At the same time,the administration group could promote cell apoptosis,the difference was statistically significant(P<0.01).Conclusion:CAIs’NeiyiⅡPrescription can play a therapeutic effect by increasing the expression level of miR-141-3p in EMS endometrial stromal cells,inhibiting the activity of downstream TGF-β1/Smad2 signaling pathway,that promoting cell apoptosis.
