摘 要:目的:研究益肾化瘀方对血管紧张素Ⅱ(AngⅡ)诱导正常人近端肾小管上皮细胞(HK-2)保护作用的可能机制。方法:HK-2细胞分为正常对照组、AngⅡ组、缬沙坦含药血清组和5%、10%、20%益肾化瘀方含药血清组,分别于培养第12、24、48小时观察细胞形态,MTT法检测细胞增殖情况,免疫荧光法检测纤连蛋白(FN)的表达,PCR法检测基质金属蛋白酶1(MMP-1)、金属蛋白酶组织抑制剂1(TIMP-1)mRNA的表达,Western blot法检测酪氨酸蛋白激酶2(JAK2)、信号转导与转录活化因子3(STAT3)、细胞因子信号传导抑制因子(SOCS1)蛋白的表达。结果:与AngⅡ组同期比较,各给药组可不同程度减轻细胞纤维化,并降低FN表达水平,MMP-1、TIMP-1 mRNA表达量和JAK2、STAT3蛋白的表达(P<0.05),并升高SOCS1蛋白表达(P<0.05),以20%益肾化瘀方含药血清组和缬沙坦含药血清组最为明显。结论:益肾化瘀方可能是通过调控JAK/STAT/SOCS1信号通路,提高抗炎水平,减少细胞外基质(ECM)沉积,对HK-2细胞起到保护作用。Objective:To study the possible mechanism of Yishen Huayu Recipe on the protective effect of AngⅡinduced HK-2 cells.Methods:HK-2 cells were divided into normal control group,AngⅡgroup,valsartan containing serum group and 5%,10%,20%Yishen Huayu Recipe containing serum groups.Cell morphology was observed by microscopy,cell proliferation was detected by MTT assay,the expression of FN was detected by immunofluorescence assay,the mRNA expression of MMP-1 and TIMP-1 were detected by PCR,and the protein expression of JAK2,STAT3 and SOCS1 were detected by Western blot at 12,24,48 hours of incubation.Results:Compared with AngⅡgroup,at the same time point,each administration group could alleviate cell fibrosis to different degrees,and reduce the expression level of FN,MMP-1,TIMP-1 mRNA and JAK2,STAT3 protein expression(P<0.05),and the expression of SOCS1 protein was increased(P<0.05),with the 20%Yishen Huayu Recipe containing serum group and valsartan containing serum group being the most obvious.Conclusion:Yishen Huayu Recipe may protect HK-2 cells by regulating the JAK/STAT/SOCS1 signaling pathway,increasing the level of anti-inflammatory and reducing ECM deposition.
