摘 要:为了探究DUR3家族基因在槟榔尿素转运过程中的作用,本研究利用拟南芥DUR3同源蛋白为探针序列,从槟榔基因组数据库中鉴定DUR3家族基因,利用生物信息学方法对其基因结构、进化关系、保守结构域、跨膜结构域和表达模式进行分析。结果表明,从槟榔基因组中共鉴定出2个DUR3基因,与其他植物的DUR3同源蛋白类似,具有15个跨膜结构域,其保守结构域和基序位置也高度相似。不同物种的DUR3氨基酸序列构建的进化树显示,双子叶植物和禾本科单子叶植物各聚为一支,而棕榈科单子叶的植物单独聚为一支,且与双子叶植物亲缘关系更近。DUR3直系同源基因的Ka/Ks比值小于1,这表明这些物种的DUR3基因在物种进化中受到纯化选择压力。本研究为深入研究槟榔的DUR3基因的功能提供了理论依据。In order to explore the role of DUR3 family genes in the transport of urea by A. catechu, we used the Arabidopsis thaliana DUR3 homologous protein as a probe sequence to identify DUR3 family genes from the A.catechu genome database in this paper. The gene structure, evolutionary relationship, conserved domains, motif distribution, transmembrane domains and expression patterns were analyzed using bioinformatics methods. The results showed that a total of two DUR3 genes were identified from the A. catechu genome, similar to DUR3 homologous proteins of other plants, with 15 transmembrane domains, and their conserved domains and motif positions were also highly similar. The phylogenetic tree constructed from the DUR3 amino acid sequences of different species showed that dicotyledonous plants and gramineous monocotyledonous plants each clustered into a branch,while palm monocotyledonous plants clustered into a single branch and were more closely related to dicotyledonous plants. The Ka/Ksratios of DUR3 orthologs were less than 1, suggesting that DUR3 genes in these species were under purifying selection pressure during species evolution. This study provides a theoretical basis for in-depth study of the function of DUR3 gene of A. catechu.
