摘 要:为建立尾叶桉(Eucalyptus urophylla) GLU4遗传转化体系,本研究探寻了不同外植体类型、生长调节剂对愈伤诱导及不定芽再生的影响。结果发现茎段最适宜作为外植体,改良MS培养基+1.5 mg/L NAA+1.5 mg/L 2,4-D+1.0 mg/L 6-BA为最佳愈伤诱导培养基,诱导率达到86.00%,愈伤组织在改良MS培养基+2.0 mg/L 6-BA+1.0 mg/L NAA中获得6.67%的最高分化率。利用建立的遗传转化体系将携带EuCAD基因全长RNAi片段的p CAD-RNAi载体转化GLU4,获得26个转化株系,其中21个株系经鉴定为阳性,阳性率80.77%。尾叶桉GLU4是中国南方广泛使用的优良纸浆材人工林树种,本研究建立的GLU4遗传转化体系将有助于推进尾叶桉基因工程育种进程,为尾叶桉木质素定向调控育种提供技术参考。In order to establish genetic transformation system of Eucalyptus urophylla clone GLU4, the effects of different explant and growth regulators on callus induction and shoot regeneration of were studied. The results showed that the stem segment was the most suitable explant. The best medium for callus induction was improved MS medium+1.5 mg/L NAA+1.5 mg/L 2,4-D+1.0 mg/L 6-BA, inwhich 86.00% callus induction rate was obtained.The highest regeneration rate of callus was 6.67% in improved MS medium+2.0 mg/L 6-BA+1.0 mg/L NAA. The p CAD-RNAi vector that carrying the full-length RNAi fragment of EuCAD gene was transformed into clone GLU4using the established genetic transformation system. 26 transformed lines were obtained, of which 21 lines were identified as positive, with a positive rate of 80.77%. GLU4 is an excellent pulp wood plantation species widely used in southern China. The genetic transformation system of GLU4 established in this study will help to promote the genetic engineering breeding process of E. urophylla, and provide technical reference for lignin oriented regulation breeding of E. urophylla.
