摘 要:目的 观察内源性大麻素单酰甘油脂肪酶抑制剂对心肺复苏后大鼠肾脏血栓素B2(TXB2)、前列腺素E2(PGE2)和前列腺素D2(PGD2)的影响。方法 将健康雄性SD清洁级大鼠分为3组,Sham组、CPR组和CPR+URB602组。CPR+URB602组和CPR组建立心肺复苏模型,并分别在复苏后即刻给予肾上腺素,自主循环恢复(ROSC)后1 min腹腔注射URB602(5 mg/kg)和同等剂量溶剂。ROSC后6 h后取标本检测肾脏组织TXB2、PGE2和PGD2、内源性大麻素2-酰基甘油(2-AG)、N-花生四烯酸氨基乙醇(AEA)、花生四烯酸(AA)含量。结果 CPR+URB602组和CPR组在ROSC后6 h AEA、2-AG、AA、PGE2、PGD2和TXB2都升高,CPR组AA、PGE2、PGD2和TXB2较CPR+URB602组升高更明显(P<0.05),但CPR+URB602组2-AG升高较CPR组更明显(P<0.05)。结论 心搏骤停-心肺复苏后肾脏内源性大麻素系统和花生四烯酸代谢通路激活,而内源性大麻素单酰甘油脂肪酶水解酶抑制剂URB602可以抑制AA代谢。Objective To observe the effect of endocannabinoid mono-glycerolipase(MAGL) inhibitor on the levels of TXB2,PGE2 and PGD2 in rat kidney after cardiopulmonary resuscitation(CPR).Methods Healthy male SD rats were divided into 3 groups: Sham group, CPR group and CPR + URB602 group. CPR + URB602 and CPR groups were establishing CPR models and epinephrine was given immediately after resuscitation. URB602(5 mg/kg) and an equivalent dose of solvent were intraperitoneally injected one minute after spontaneous circulation recovery(ROSC). The contents of TXB2,PGE2,PGD2,endocannabinoid 2-acylglycerol(2-AG),N-arachidonic acid aminoethanol(AEA),arachidonic acid (AA) content in kidneys were measured 6 hours after ROSC.Results The levels of AEA,2-AG,AA,PGE2,PGD2,and TXB2 were all increased in CPR+URB602 group and CPR group at 6 hours after ROSC. Compared with CPR+URB602 group, the AA,PGE2,PGD2,and TXB2 increased significantly(P<0.05),but the increase of 2-AG in CPR+URB602 group was more obvious than that in CPR group(P<0.05).Conclusion Cardiac arrest-CPR activates the renal endocannabinoid system and arachidonic acid metabolic pathway activation, whereas the endogenous cannabinoid mono-glycerol lipase(MAGL) hydrolase inhibitor URB602 inhibits arachidonic acid metabolites.
