摘 要:以‘长富2’苹果片红芽变的果皮为试材,克隆了溶质转运蛋白家族基因Md SLC35F2-like(Solute Carrier Family 35 Member F2-like)的开放阅读框(ORF),其长度为1041 bp,编码346个氨基酸。利用在线软件进行生物信息分析,该蛋白是1个稳定的疏水性α螺旋跨膜蛋白,定位于细胞膜。同源性分析表明,Md SLC35F2-like与白梨的Pb SLC35F1-like同源性最高;系统进化分析表明,其与白梨的进化亲缘关系最近,与可可、巨桉的进化亲缘关系最远。分析‘长富2号’及其片红芽变果实着色期花青苷的积累与Md SLC35F2-like表达量的变化,结果表明,摘袋后Md SLC35F2-like表达量在‘长富2号’及其2个片红芽变果实果皮中均呈先升高后降低的趋势,在摘袋后3d表达量最高,而后逐渐降低,2个芽变材料均显著高于其野生型,表达量与花青苷含量呈正相关关系。在其他颜色品种上进行验证分析,Md SLC35F2-like在红色果皮品种‘烟富3号’果皮中的表达量极显著高于绿色果皮品种‘金陵’,在红色果肉品种‘红色之爱’果肉中的表达量显著高于白肉品种‘烟农早富’。以上结果进一步证明Md SLC35F2-like与花青苷积累有关。Anthocyanin is the main pigment affecting the skin color of apple fruit.In our previous study,Md SLC35F2-like was expressed at higher levels in the‘Nagafu 2’apple red bud mutant than in the wild-type.The open reading frame(ORF)of Md SLC35F2-like was cloned,it is 1041bp,encoding 346 amino acids,which was predicted to be a hydrophobicαhelix transmembrane protein with the highest homology to the Pb SLC35F1-like of Pyrus×bretschneideri.Phylogenetic analysis showed that it had the closest evolutionary relationship with P.×bretschneideri,and the furthest evolutionary relationship with Theobroma cacao and Eucalyptus grandis.Md SLC35F2-like sequence and its expression rules before and after the fruit bagging were analyzed.After unbagging,the expression level of Md SLC35F2-like increased firstly,and then decreased with a peak on the third day.The expression level of Md SLC35F2-like in the two kinds of buds was significantly higher than that of‘Nagafu 2’.The above results indicated that the expression level of Md SLC35F2-like gene is positively correlated with anthocyanine content.Meanwhile,its expression levels in different color apple cultivars were compared.The expression of Md SLC35F2-like in red peel cultivars was significantly higher than that in green peel cultivars,and the expression level in red pulp cultivars was higher than that in non-red pulp cultivars.It suggests that Md SLC35F2-like may participate in the anthocyanine accumulation process.
