月泰贴脐片影响宫寒血瘀型肾虚不孕大鼠生育功能实验研究

摘　　要：目的:观察月泰贴脐片对宫寒血瘀型肾虚不孕大鼠生育功能的影响。方法:将60只SD雌性大鼠随机分为:正常组、模型组、高剂量组[0.99mg/(kg·日)],中剂量组[0.495mg/(kg·日)]、低剂量组[0.2475mg/(kg·日)],每组12只大鼠。通过灌胃给予羟基脲(450 mg/kg) 15 d,建立大鼠宫寒血瘀肾虚不孕模型。将月泰贴脐片按剂量贴于大鼠肚脐(神阙穴)。第11天,每组雌鼠和雄鼠2:1自由关笼4 d受孕。第15天,将笼中的雄性大鼠取出。7 d后处死雌性大鼠,记录孕鼠数量。采集血清样本,测定血清促排卵激素(FSH)、黄体生成素(LH)和雌二醇(E2)。HE染色观察子宫形态变化。结果:月泰贴脐片高剂量组大鼠血液中FSH、 LH、 E2的水平高于模型组(P<0.05);月泰贴脐片高剂量组子宫内膜卵泡数目增多,子宫内膜和腺体发育较好。结论:月泰贴脐片可以改善宫寒血瘀型肾虚不孕大鼠症状,并提高大鼠妊娠率,促进子宫内膜和腺体发育。Objective To observe the effect of Yuetai paste navel tablet on the reproductive function of infertile rats with kidney deficiency due to uterine cold and blood stasis.Methods 60 female SD rats were randomly divided into normal group, model group,high dose group [0. 99 mg/(kg·day)], middle dose group [0. 495mg/(kg ·day)] and low dose group [0. 2475mg/(kg·day)], with 12 rats in each group). The infertility model of uterine cold, blood stasis and kidney deficiency in rats was established by intragastric administration of hydroxyurea(450mg/kg) for 15 days. Yuetai paste navel tablets were affixed to the navel of rats(Shenque acupoint) according to the dose. On the 11th day, each group of female and male rats were freely closed at 2: 1 and conceived for4 days. On the 15th day, the male rats in the cage were taken out. After 7 days, the female rats were killed and the number of pregnant rats was recorded. Serum samples were collected and ovulation stimulating Follicle-Stimulating Hormone(FSH), Luteinizing Hormone(LH) and Estradiol(E2) were measured. The morphological changes of uterus were observed by HE staining.Results The levels of FSH, LH and E2 in the blood of rats in the high dose Yuetai paste umbilical tablet group were higher than those in the model group, and the number of endometrial follicles increased and the endometrium and glands developed better in the high dose Yuetai paste umbilical tablet group.Conclusion Yuetai paste navel tablet can enhance the symptoms of infertile rats with kidney-deficiency due to uterine cold and blood stasis, increase the pregnancy rate, and promote the development of endometrium and glands.
