裸花紫珠总黄酮提取工艺及抗HBV活性成分研究

摘　　要：目的:探讨裸花紫珠总黄酮的提取工艺及抗HBV活性,为其深入研究提供参考依据。方法:用正交试验设计优选总黄酮的最佳提取工艺;用MTT法测试总黄酮提取物对HepG2.2.15细胞抑制作用。结果:裸花紫珠总黄酮的最佳提取工艺为:60%乙醇回流提取2次,每次2 h,料液比为1∶20,测得提取物总黄酮百分含量为11.29%;总黄酮提取物对HepG2.2.15细胞分泌的表面抗原HBsAg和e-抗原HBeAg有抑制作用,同时对HBV-DNA的复制有一定的抑制作用。结论:裸花紫珠总黄酮提取物具有一定的抗HBV活性,黄酮类化合物可能为抗HBV活性成分。Objective To investigate the extraction process and anti-HBV activity of total flavone from Callicarpa nudiflora,and provide a reference for its further research.Methods The best extraction process was optimized by orthogonal test,and The inhibitory effects of the total flavone extract on HepG2.2.15 was tested by MTT method.Results The best extraction process of total flavone from C.nudiflora was 60%ethanol with material-liquid ratio 1:20 to reflux extract 2 times,2 hours each time,and the total flavone content of the extract was 11.29%.The total flavone extract had definite inhibitory effects on HBsAg,HBeAg and HBV-DNA of HepG2.2.15 cell.Conclusion The total flavone extract from C.nudiflora has a certain anti-HBV activity,and the flavones may be the active component.
