益智方水提物及其不同萃取部位对小鼠学习记忆障碍的改善作用

摘  要:目的:观察益智方水提物及其不同萃取部位对东莨菪碱致小鼠记忆障碍的改善作用。方法:选取SPF级小鼠80只,随机分为对照组、模型组、阳性组、石油醚组、乙酸乙酯组、正丁醇组、多糖组、水煎液组,每组10只。对照组和模型组小鼠予生理盐水灌胃,阳性组予吡拉西坦灌胃,其余各组小鼠予益智方水提物相应不同萃取部位灌胃,灌胃体积0.1 mL/10 g,每天固定时间给药1次,连续7 d。除对照组外的小鼠腹腔注射氢溴酸东莨菪碱(2 mg/kg)制备小鼠学习记忆障碍模型。应用Morris水迷宫实验、跳台实验观察益智方不同组分对小鼠学习记忆能力的改善作用,HE染色小鼠脑组织观察海马组织神经元的变化。结果:跳台实验中,益智方不同组分均能使学习记忆障碍小鼠的错误次数减少,跳台潜伏期延长;Morris水迷宫实验中,益智方不同组分均能使小鼠的逃避潜伏期延长,穿越平台次数增加;其中石油醚的作用最为显著,石油醚组分作为益智方有效部位可保护小鼠海马神经元形态,降低神经元受损程度。结论:益智方水提物及其不同萃取部位均能不同程度改善东莨菪碱致小鼠学习记忆能力,其中石油醚组分最为明显,乙酸乙酯组分、水煎液和多糖组分次之,正丁醇组分相对不明显。石油醚组分作为益智方有效部位可有效保护小鼠海马组织。Objective To observe the effect of water extract of Yizhi prescription and its different extraction parts on memory impairment induced by scopolamine in mice. Methods 80 SPF mice were randomly divided into control group, model group, positive drug group, petroleum ether group, ethyl acetate group, n-butanol group, polysaccharide group and decoction group.The mice in the control group and the model group were treated with ig normal saline, the positive group was treated with ig piracetam, and the mice in the other groups were ip reated with Yizhi prescription water extract corresponding to different extraction parts, with a gavage volume of 0.1 mL/10 g, once a day at a fixed time for 7 days.Except for the control group, the model of learning and memory impairment in mice was prepared by scopolamine hydrobromide(2 mg/kg).Morris water maze test and platform jumping test were used to observe the improvement of different components of Yizhi prescription on learning and memory ability of mice. HE staining was used to observe the changes of neurons in hippocampus of mice. Results In the platform jumping experiment, different components of Yizhi prescription can reduce the number of errors and prolong the platform jumping latency of learning and memory impaired mice;In Morris water maze experiment, different components of Yizhi prescription can prolong the escape latency and increase the number of crossing the platform;Petroleum ether has the most significant effect. As an effective part of Yizhi prescription, petroleum ether can protect the morphology of hippocampal neurons and reduce the degree of neuronal damage.Conclusions The water extract of Yizhi prescription and its different extraction parts can improve the learning and memory ability of mice induced by scopolamine to varying degrees, among which the petroleum ether component is the most obvious, the ethyl acetate component, Decoction and polysaccharide component are the second, and the n-butanol component is not obvious.As an effective part of Yizhi prescriptio
