摘 要:目的:研究苦豆子总碱(TASA)对大鼠慢性酒精性肝损伤的保护作用和作用机制。方法:雄性大鼠随机分为空白组,模型组,TASA 14 mg/kg、28 mg/kg和56 mg/kg组,联苯双酯组(2.4 mg/kg),连续灌胃给药8周。同时除空白组外,其它组每天灌胃56°白酒,每周递增,连续8周。末次给药后,取血测ALT、AST、MDA、SOD、TNF-α、IL-6、TGF-β1和IFN-γ各项指标,并进行肝脏组织病理学检查。结果:与空白组相比,模型组各项指标均发生显著变化,与模型组相比,TASA可显著降低ALT、AST、MDA、IL-6、TGF-β1和IFN-γ,显著升高SOD活性,组织病理切片显示显著减轻炎症细胞浸润。结论:说明TASA对慢性酒精性肝损伤具有很好的保护作用,可能与其抗氧化应激和抗炎作用以及促进肝细胞再生、阻止纤维化进程有关。Objective To study the protective effect and mechanism of total alkaloid of sophora alopecuroides(TASA)on chronic alcoholic liver injury in rats.Methods Male rats were randomly divided into blank group,model group,TASA 14,28,56 mg/kg group and bifendate group(2.4 mg/kg),and were given intragastrically for 8 weeks.At the same time,except for the blank group,the other groups were given 56%liquor every day for 8 consecutive weeks,increasing every week.After the last administration,blood was taken to measure ALT,AST,MDA,SOD,TNF-α,IL-6,TGF-β1 and IFN-γ,and histopathological examination of the liver was performed.Results Compared with the blank group,all indicators of the model group changed significantly;compared with the model group,TASA can significantly reduce ALT,AST,MDA,IL-6,TGF-β1 and IFN-γ,and significantly increase SOD activity;histopathological sections show a significant reduction in inflammatory cell infiltration.Conclusions It shows that TASA has a good protective effect on chronic alcoholic liver injury,which may be related to its anti-oxidative stress and anti-inflammatory effects,promote liver cell regeneration and prevent the process of fibrosis.
