多指标权重分析法结合正交试验优选罗布麻冲剂的水提工艺

摘　　要：目的:优化罗布麻冲剂的水提工艺并确定其参数。方法:以金丝桃苷转移率、总黄酮含量以及出膏率为评价指标,采用高效液相色谱法和紫外分光光度计法测定指标含量,AHP-CRITIC混合加权法确定各指标权重系数,设计L_(9)(3^(4))正交试验筛选罗布麻叶冲剂的水提工艺中提取次数、提取时间和加水倍量,并进行验证试验。结果:AHP-CRIT-IC混合加权法确定金丝桃苷转移率、总黄酮的含量以及出膏率的权重分别为0.4768,0.4346和0.0886,筛选的提取工艺为罗布麻叶加10倍量水、加热回流3次,每次0.5 h。3次验证试验结果显示,金丝桃苷的提取率为90.29%,总黄酮的含量为105.84 mg/g,出膏率为29.87%,综合评分为98.44(RSD=1.86%,n=3)。结论:优选的罗布麻冲剂水提工艺重复性好、稳定可行,可为罗布麻冲剂的工业化生产提供参考。Objictive To optimize the water extraction process of Apocynum granules and determine its parameters.Methods Taking the transfer rate of hypericin,the content of total flavonoids and the extract rate as indicators,the contents of indicators were determined by HPLC and ultraviolet spectrophotometer,and the weight of each indicator was determined by AHP-CRITIC mixed weighting method.The L_(9)(3^(4))orthogonal test was designed to screen the parameters in the water extraction process of Apocynum granules:extraction times,extraction time and water addition amount,and the verification test was carried out.Results The AHP-CRITIC mixed weighting method was used to determine the weight coefficients of the transfer rate of hypericin,the content of total flavonoids and the extract rate,which were 0.4768,0.4346 and 0.0886,the extraction process was adding 10 times of water and heating reflux 3 times,0.5h each time.The results of the three verification tests showed that the extraction rate of hyperin was 90.29%,the content of total flavonoids was 105.84 mg/g,the dry paste yield was 29.87%,and the average comprehensive score was 98.44(RSD=1.86%,n=3).Conclusion The optimal water extraction process of apocynum granules has good repeatability,stability and feasibility,which can provide a reference for the industrial production of apocynum granules.
