摘 要:为探究丹参多糖(SMPs)在奶牛乳腺炎中的应用价值,本研究以分娩后7 d小鼠为对象,SMPs溶液灌胃1周后使用脂多糖(LPS)构建小鼠乳腺炎的模型,24 h后处死小鼠收集乳腺组织检测。H&E结果表明,脂多糖会诱导乳腺组织发生严重的炎症反应;ELISA结果显示,SMPs用药组中TNF-α,IL-6和IL-1β在乳腺组织表达水平显著下降。Western Blot结果显示,SMPs用药组中TLR4,MyD88和NF-κB蛋白表达水平显著降低。RT-qPCR结果显示,SMPs用药组中TNF-α,IL-6和IL-1βmRNA转录水平显著降低。结果表明,SMPs对LPS诱导的乳腺炎症具有保护作用,其抗炎作用与抑制TLR4信号通路中关键蛋白表达水平有关。In order to explore the application value of Salvia miltiorrhiza polysaccharides(SMPs)in dairy mastitis,mice after 7 days delivery were used as subjects in this study.Mouse mastitis model was established using lipopolysaccharide(LPS)after intragastric administration of SMPs solution for 1 week,and mice mammary tissues were collected after sacrifice at 24h of LPS treatment.H&E results showed that LPS could induce severe inflammatory response in breast tissue.ELISA results showed that the expression levels of TNF-α,IL-6 and IL-1β significantly decreased in breast tissue of the SMPs treatment group.Western Blot results showed that TLR4,MyD88 and NF-κB protein levels significantly decreased in SMPs treatment group.RT-qPCR results further showed the significant reduces of the mRNA transcription levels of TNF-α,IL-6 and IL-1β in the SMPs treatment group.The results showed that SMPs had a protective effect on LPS-induced mammary inflammation,and its anti-inflammatory effect was related to the inhibition of the expression of key proteins in the TLR4 signaling pathway.
