摘  要:目的 运用网络药理学和生物信息学的方法探讨补中益气汤治疗气虚发热证的机制。方法 使用中药系统药理数据库(TCMSP)以补中益气汤的8味中药作为关键词搜索其主要化学成分,同时筛选作用靶点并利用Uniprot数据库完成蛋白靶点对照,通过用genecard数据库获取气虚发热证即与发热相关的疾病靶基因信息,将初步筛选后的成分相关靶点和疾病靶基因制作韦恩图,获得补中益气汤有效成分与疾病的交集靶点;通过STRING平台和Cytoscape3.7.2软件筛选关键靶点并构建PPI图及中药-有效化学成分-疾病靶点网络图;以DAVID完成靶基因的GO功能分析及KEGG通路分析并绘制条形图。结果 TCMSP数据库获得补中益气汤的主要有效化学成分共153个,分别是黄芪有16个,升麻有17个,人参有20个,陈皮有5个,甘草有74个有效化学成分,柴胡有14个有效化学成分。使用genecard疾病数据库筛选出发热相关靶基因1 501个;其中药物靶点和疾病靶点治疗发热的交集靶点105个。交集靶点蛋白-蛋白之间相互作用网络中节点连接度较高的靶点有肿瘤坏死因子(TNF)、白介素(IL6)、AKT1、JUN、VEGFA。结论 补中益气汤治疗气虚发热证具有多成分、多靶点、多通路特征,从整体角度阐述了中医药有效的作用机制,为进一步研究提供分子层面的思路和依据。Objective To explore the mechanism of Buzhong Yiqi Decoction for Spleen-Qi Deficiency with fever symptoms based on network pharmacology and bioinformatics.Methods Main active components was searched using 8 herbs name of Buzhong Yiqi Decoction as the key words via Traditional Chinese Medicine Systems Pharmacology(TCMSP);meantime, the action target was screened, Protein-protein interactions were performed in Universal Protein Resource(UniProt) database. Genecard database was used to obtain the target gene of fever-related diseases. The Venn diagram was set up based on the primary result to screen the drug-disease intersection targets corresponding 8 herbs in Buzhong Yiqi Decoction.The key targets were screened by the STRING platform and Cytoscape3.7.2 software to construct protein-protein interaction(PPI) network and Chinese medicine-active component-disease target network. target genes of the Gene Ontology(GO) analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway were conducted by the Database for Annotation, Visualization, and Integrated Discovery(DAVID), and the bar graphs were drawn.Results A total of 153 main active components of Buzhong Yiqi Decoction were obtained via TCMSP database, includeing Huangqi for 16, Shengma for 17, Renshen for 20, Chenpi for 5, Gancao for 74. 105 drug-disease intersection targets were obtained among 1501 fever-related target genes screened from Genecard database. The targets with higher node connectivity in the PPI network of intersecting targets were tumor necrosis factor(TNF), interleukin-6(IL-6), AKT1, JUN and vascular endothelial growth factor A(VEGFA).Conclusion This study has comprehensively verified the mechanism of Buzhong Yiqi Decoction in the treatment of Spleen Qi Deficiency with fever syndrome, characteristed by multi-component, multi-target and multi-channel, providing new evidence and research ideas for future research.
