摘 要:选取从河南棉花上分离的尖孢镰刀菌(Fusarium oxyspomm f.sp.vasinfectum)异核体菌株Ag149及其两个表型差异显著的同核型分离子Agl49-Ⅰ和Ag149-Ⅲ为材料,对它们的核糖体基因ITs区段进行测序分析。结果表明,供试菌株核糖体基因ITS区段碱基组成完全相同,与GenBank中镰刀菌的ITs区序列进行同源性比较发现,属内种间的同源性在34%~99%之间,而大部分尖孢镰刀菌种内不同菌株之间的同源性均在98%以上。序列分析表明,ITS区对于镰刀菌属内种间的差异鉴别具有参考价值,但看来并不适合尖孢镰刀菌种内或种以下水平的鉴定和分类。Two segregants (Agl49- Ⅰ and Ag149-Ⅲ) were isolated from a heterokaryon of Fusarium oxysporum f.sp. vasinfectum Agl49. They were different in some phenotypic characters. Their internal transcribed spacer (ITS) region in rRNA genes were sequenced, and showed no difference among the three strains. Comparing their ITS sequences with related strains in GenBank, the homology of ITS among Fusarium species were 34% to 99%, while the homology of ITS among Fusarium oxysporum strains were more than 98%.The ITS sequence might be used for the differential identification of species of Fusarium, but could not be used for the identification of subspecies or biotypes of Fusaium oxysporum.
