摘 要:目的:研究蒙药音达日-4味汤散乙酸乙酯提取物的解热镇痛作用及对临床常见致病菌的体外抗菌作用。方法:利用背部皮下注射干酵母法建立小鼠发热模型,通过测量其2 h、4 h、6 h、8 h等时间点体温,观察音达日-4味汤提取物对发热的影响;通过热板法测出小鼠痛阈值,观察音达日-4味汤散对镇痛反应的影响;分别采用琼脂平板法及微孔板TTC法测定音达日-4味汤提取物对金黄色葡萄球菌等11种临床常见致病菌的最低抑菌、杀菌浓度。结果:干酵母致小鼠发热实验结果表明,与模型组比较,音达日-4味汤散乙酸乙酯提取物中(42 g·(kg·d)^(-1))(P<0.01)、高(84 g·(kg·d)^(-1))(P<0.01)剂量组小鼠体温分别在造模后8 h、4 h差异最为显著,其解热作用可持续8 h以上;热板致小鼠镇痛实验发现,与模型组比较,蒙药音达日-4味汤乙酸乙酯提取物中(42 g·(kg·d)^(-1))(P<0.05)、高(84 g·(kg·d)^(-1))(P<0.01)剂量组痛阈值均有明显升高,痛反应潜伏期有所延长;此外,音达日-4味汤散乙酸乙酯提取物对金黄色葡萄球菌等11种临床常见耐药致病菌均表现出了较为普遍的抑菌和杀菌作用。结论:蒙药音达日-4味汤散乙酸乙酯提取物具有较明显的解热及镇痛作用;初步判断乙酸乙酯提取物为音达日-4味汤散解热、镇痛及抗菌作用的有效部位,对金黄色葡萄球菌和肠球菌抑制能力最强。Objective:To study the antipyretic and analgesic effects of ethyl acetate extract from Yindari-4 decoction power and its antibacterial effects on clinical common pathogens in vitro.Methods:The mouse fever model was established by subcutaneous injection of dry yeast into the back.The effect of Yindari-4 decoction extract on fever was observed by measuring the mice temperature of 2 h,4 h,6 h,8 h and other time points.The pain threshold of mice was measured by hot plate method,and the effect of Yindari-4 decoction powder on analgesic reaction was observed.The minimum inhibitory and minimum bactericidal concentration of Yindari-4 decoction extract against 11 clinical common pathogens such as Staphylococcus aureus were determined by agar plate method and microporous plate TTC method respectively.Results:The experimental results of dry yeast-induced fever in mice showed that compared with the model group,the body temperature of mice in the middle dose group(42 g·(kg·d)^(-1))(P<0.01)and high dose group(84 g·(kg·d)^(-1))(P<0.01)of the ethyl acetate extract from Yindari-4 decoction powder had the most significant difference at 8 h and 4 h after modeling,respectively,and its antipyretic effect lasted for more than 8 h.The analgesic experiment induced by hot plate in mice showed that compared with the model group,the pain threshold in the middle dose group(42 g·(kg·d)^(-1))(P<0.05)and high dose group(84 g·(kg·d)^(-1))(P<0.01)of Mongolian medicine Yindari-4 decoction ethyl acetate extract were significantly increased,and the latency of pain reaction was prolonged.In addition,the ethyl acetate extract of Yindari-4 decoction powder showed general bacteriostatic and bactericidal effects on 11clinically ethyl acetate common drug-resistant pathogens such as Staphylococcus aureus.Conclusion:The ethyl acetate extract of Mongolian medicine Yindari-4 decoction powder has obvious antipyretic and analgesic effects.It is preliminarily concluded that the ethyl acetate extract is the effective part of Yindari-4 decoction powder in
