KIF18B在高级别卵巢浆液性癌中表达及与免疫细胞浸润的相关性研究

摘　　要：目的 研究KIF18B在高级别卵巢浆液性癌中的表达情况,分析其与预后及免疫细胞浸润的关系。方法 本研究共纳入105例高级别卵巢浆液性癌患者,实时定量PCR及免疫组织化学法分析KIF18B在105对癌与癌旁组织中mRNA及蛋白的表达水平,并分析其与高级别卵巢浆液性癌患者预后的关系。采用TIMER网站分析KIF18B表达与免疫细胞浸润的相关性,同时采用实时定量PCR验证KIF18B表达水平与免疫浸润细胞标志分子的相关性。结果 KIF18B的表达在卵巢浆液性癌与正常卵巢组织中存在明显差异,与癌旁组织相比,KIF18B mRNA水平在癌组织中明显增高;KIF18B蛋白水平在卵巢癌组织中亦明显高于癌旁正常组织。COX回归分析显示,高表达KIF18B蛋白的卵巢癌患者具有更短的生存期。TIMER数据库显示KIF18B基因与免疫细胞浸润相关,笔者的研究发现KIF18B的表达与Treg标记物(FOXP3,CCR8,STAT5B及TGFB1)相关。此外,它还与多种其他免疫细胞标志物相关。结论 高级别卵巢浆液性癌组织中KIF18B蛋白高表达与患者的不良预后密切相关,且可作为肿瘤免疫细胞浸润的重要标志物,是评估卵巢浆液性癌患者预后及免疫浸润的重要标记物。Objective To study the expression of KIF18 B in high-grade ovarian slurry cancer, analyze its relationship with prognosis and immune cell infiltration.Methods In this study, a total of one hundred and five patients with high-grade ovarian slurry cancer were included in real-time quantitative PCR and immunohistochemical chemistry analysis.KIF18 B’s expression of MRNA and protein in 105 cancers and cancer tissues, and analyzed its high-level ovarian slurry Prognosis of patients with sex cancer.The Timer website analyzes the correlation between KIF18 B expression and immune cell infiltration, and the correlation between real-time quantitative PCR verification KIF18 B expression level and immunohistocyte logo molecule.Results The expression of KIF18 B has a significant difference in ovarian slurry cancer and normal ovarian tissue.Compared with the tissue of the cancer, the KIF18 B mRNA level is significantly higher in the cancer tissue;the KIF18 B protein level is also significantly higher than the cancer tissue than the cancer tissue.Normally organize.COX regression analysis shows that patients with ovarian cancer expressing KIF18 B protein have shorter survival period.The Timer database shows that the KIF18 B gene is related to the infiltration of immune cells.I have found that the expression of KIF18 B is related to the TREG marker(Foxp3,CCR8,STAT5 B and TGFB1).Inaddition, it is related to a variety of other immune cells.Conclusion KIF18 B of high-grade ovarian slurry cancer tissue is closely related to patients’ adverse prognosis, and can be used as an important symbol of infiltration of tumor immune cells.It is an important marker for evaluating the prognosis and immune invasion of patients with ovarian slurry cancer.
