摘  要:目的:探索低氧诱导因子(HIF)-1α、HIF-2α在先天性小耳畸形耳软骨与正常耳软骨中的表达,并进行差异分析。方法:选取单侧先天性小耳畸形患者的耳软骨7例为实验组,正常耳软骨4例为对照组,提取软骨组织总RNA和总蛋白,分别采用实时荧光定量聚合酶链反应(RT-qPCR)方法和蛋白质印迹法(Western blot)方法检测HIF-1α、HIF-2α在mRNA水平和蛋白质水平的表达情况,并应用免疫组织化学染色法检测HIF-1α、HIF-2α在组织中的定位,两组之间比较采用独立样本t检验。结果:RT-qPCR显示实验组中HIF-1α相对表达量低于对照组(0.256±0.097比0.393±0.052,t=2.566,P<0.05),实验组HIF-2α相对表达量低于对照组(0.230±0.134比0.244±0.131),且HIF-1α差异有统计学意义。Western blot显示HIF-1α在实验组中的相对表达量低于对照组(0.242±0.034比1.119±0.085,t=19.314,P<0.05),HIF-2α在实验组中的相对表达量低于对照组(0.388±0.067比1.129±0.089,t=14.409,P<0.05),差异有统计学意义。结论:HIF-1α、HIF-2α在先天性小耳畸形及正常耳软骨中均有表达,主要位于细胞核内,且在mRNA和蛋白质水平上小耳畸形耳软骨中的表达量低于正常耳软骨。Objective To explore the expression of hypoxia-inducible factor(HIF)-1α,HIF-2αin the congenital microtia cartilage and the normal ear cartilage,and to analyze the difference.Methods There were 7 cases of unilateral congenital microtia patients selected as the experimental group and 4 cases of normal ear cartilage as the control group.Total RNA and total protein of cartilage tissue were extracted.The mRNA and protein expressions of HIF-1αand HIF-2αwere detected respectively by real-time fluorescence quantitative polymerase chain reaction(RT-qPCR)and Western blotting,and the location of HIF-1αand HIF-2αin cartilage tissues was detected by immunohistochemical staining.Two-tailed student’s t-test was used for comparisons between the two groups.Results RT-qPCR showed the relative expression of HIF-1αwere lower in the congenital microtia than in the normal auricle cartilage(0.256±0.097 vs.0.393±0.052,t=2.566,P<0.05),the relative expression of HIF-2αwere lower in the congenital microtia than in the normal auricle cartilage(0.230±0.134 vs.0.244±0.131),and the difference in HIF-1αexpression was statistically significant.Western blotting showed that the relative expression of HIF-1αin microtia cartilage was lower than that in normal ear cartilage(0.242±0.034 vs.1.119±0.085,t=19.314,P<0.05),and the relative expression of HIF-2αin microtia cartilage was lower than that in normal ear cartilage(0.388±0.067 vs.1.129±0.089,t=14.409,P<0.05),and the difference was statistically significant.Conclusion Both congenital microtia and normal ear cartilage can express HIF-1αand HIF-2α,mainly in the nucleus.And HIF-1αand HIF-2αin microtia cartilage were lower than those in normal ear cartilage at the mRNA and protein levels.
