摘 要:目的分子生物学hucMSC-exo传递mir-124增进大鼠肝再生的调节作用及机制。方法采用大鼠构建动物模型,构建过表达载体及转染,采用BrdU检测及定时定量PCR。结果术后注射组和未注射组的表达关系,mir-193a、mir-204、mir-124以及不同组间mir-93、mir-10a-5p表达存在着差异,mir-124组间有明显的表达差异。结论hucMSCs衍生的外泌体mir-124通过负性调节大鼠部分肝切除模型中的Foxg1,促进肝再生并抑制肝损伤,这些发现可用于开发肝再生的新疗法。Objective To study the regulatory role and mechanism of hucMSC-exo transduction of mir-124 in promoting rat liver regeneration.Methods Rats were used to construct the animal model,and the overexpression vector was constructed and transfected.BrdU detection and timed quantitative PCR were used.Results Expression relationship between injected and non-injected groups after surgery:There were differences in the expression of mir-193a,mir-204,mir-124,mir-93,mir-10a-5p among different groups,and there was a significant difference in the expression of mir-124 among different groups.Conclusion HucMSCs-derived exosomal mir-124 promotes liver regeneration and inhibits liver injury by negatively regulating Foxg1 in a rat partial hepatectomy model.These findings can be used to develop new therapies for liver regeneration.
