摘 要:目的探讨自然杀伤细胞(NK细胞)在非创伤性股骨头坏死(non-traumatic femoral head necrosis,NONFH)患者外周血及关节液中的表型变化及RANK/RANKL/OPG表达量的变化。方法通过流式细胞术学技术对比分析两组外周血及关节液NK细胞表型变化及其表面受体HLA-DR及NKp30(CD337)表达不同,通过ELISA技术测定两组外周血及关节液中人骨保护素(osteoclastogenesis inhibitory factor,OPG)、核因子κB受体激沾因子(receptor activator of NF-κB,RANK)、核因子κB受体配体激沾因子(receptor activator of NF-κB ligand,RANKL)的表达差异。结果结果显示在NONFH患者中外周血及关节液中NK细胞数量明显增加,其中以CD3-CD56+NK为主,其表面受体HLA-DR显著上调,NKp30表达明显下调,OPG表达下调而RANK、RANKL明显上调。结论NK细胞参与NONFH疾病的进展过程,其表面受体HLA-DR增高而NKp30下调,同时OPG表达明显下调,RANK及RANKL表达上调。Objective To investigate the phenotypic changes and expression levels of RANK/RANKL/OPG of natural killer cells(NK cells)in peripheral blood and articular fluid of patients with non-traumatic femoral head necrosis(NONFH).Methods Changes of the phenotypes of NK cells and the different expression of HLA-DR and NKp30(CD337)in peripheral blood and articular fluid of the two groups were analyzed by flow cytometry.The expressions of RANK,RANKL and OPG in peripheral blood and articular fluid of the two groups were determined by ELISA.Results Results showed that the number of NK cells in peripheral blood and articular fluid of NONFH patients was significantly increased,mainly CD3-CD56+NK,and its surface receptor HLA-DR was significantly up-regulated,NKp30 expression was significantly down-regulated,OPG expression was down-regulated,and RANK and RANKL were significantly up-regulated.Conclusion NK cells are involved in the progression of NONFH disease,and its surface receptor HLA-DR is increased while NKp30 is down-regulated.Meanwhile,the expression of OPG is significantly down-regulated,and the expression of RANK and RANKL is up-regulated.
