摘  要:瘢痕是皮肤损伤愈合的必然产物,前期研究表明,沉默内源性颗粒蛋白前体(PGRN)促进胶原沉积和纤维形成。本研究继续应用小鼠皮肤孔状切除模型,在创口及其周围皮下注射Si-m-PGRN和/或LV3-TGF-β1敲减PGRN和/或TGF-β1,研究TGF-β1在PGRN调控皮肤损伤愈合纤维形成中的作用。结果显示,LV3-TGF-β1和Si-m-PGRN可以较好的敲减TGF-β1和PGRN,敲减TGF-β1逆转了PGRN对纤维形成的调控,同时逆转PGRN对下游smad3信号通路的调控。提示PGRN可通过TGF-β1调控其下游信号通路影响皮肤损伤后纤维形成。A scar is an inevitable product of skin wound healing.Previous studies have shown that silencing endogenous progranulin(PGRN)promotes collagen deposition and fibrogenesis.To study the role of TGF-β1 in PGRN regulating skin wound healing,the Si-m-PGRN and/or LV3-TGF-β1 were injected subcutaneously around the wound to knock down PGRN and/or TGF-β1,using the mouse model of skin perforation continuously.The results showed that LV3-TGF-β1 and Si-m-PGRN may knock down TGF-β1 and PGRN effectively.The knock down of TGF-β1 interfered with the effect of PGRN on fibrogenesis and the downstream smad3 signal pathway.It is suggested that PGRN regulated fibrogenesis during wound healing through the TGF-β1 signal pathway.
