摘  要:目的基于Wnt信号通路探究上调miR-184对糖尿病视网膜病变模型大鼠的干预效果。方法选取40只SD健康大鼠,10只作为正常组,剩余30只建立糖尿病视网膜病变模型,并随机分为模型组、下调miR-184组、上调miR-184组。对4组大鼠分别进行干预,同时对4组大鼠视网膜组织进行苏木素-伊红(HE)染色处理。对总胆固醇(TC)、三酰甘油(TG)、空腹血糖水平进行检测对比。采用免疫比浊法、酶联免疫吸附试验分别对谷胱甘肽(GSH)、一氧化氮(NO)、血管内皮生长因子(VEGF)、肿瘤坏死因子(TNF)-α水平进行检测。结果与正常组相比,模型组TC、TG、空腹血糖、VEGF、TNF-α、NO水平显著升高(P<0.05);与模型组相比,下调miR-184组TC、TG、空腹血糖、VEGF、TNF-α、NO水平显著升高(P<0.05);与下调miR-184组相比,上调miR-184组TC、TG、空腹血糖、VEGF、TNF-α、NO水平显著降低(P<0.05);与正常组相比,模型组GSH显著下降(P<0.05);与模型组相比,下调miR-184组GSH显著下降(P<0.05);与下调miR-184组相比,上调miR-184组GSH显著上升(P<0.05);与正常组相比,模型组糖原合成激酶(GSK)-3β表达显著升高、β-连环蛋白(catenin)、原癌基因(C-Myc)表达显著降低(P<0.05);与模型组相比,下调miR-184组GSK-3β表达显著升高、β-catenin、C-Myc表达显著降低(P<0.05);与下调miR-184组相比,上调miR-184组GSK-3β表达显著降低,β-catenin、C-Myc表达显著升高(P<0.05)。结论上调miR-184干预糖尿病视网膜病变模型大鼠,对血脂、血糖水平具有调控作用,可改善眼底视网膜环境,有效降低血管通透性,从而达到治疗糖尿病视网膜病变的效果。Objective To explore the intervention effect of up-regulating miR-184 on diabetic retinopathy model rats based on Wnt signaling pathway.Methods 40 SD healthy rats were selected,and 10 were selected as normal group,the remaining 30 were established diabetic retinopathy model,and randomly divided into model group,down-regulated miR-184 group and up-regulated miR-184 group.The four groups of rats were intervened,and the retinal tissues of the four groups of rats were treated with hematoxylin-eosin(HE)staining at the same time.The levels of total cholesterol(TC),triacylglycerol(TG)and fasting blood glucose were detected and compared.The levels of glutathione(GSH),nitric oxide(NO),vascular endothelial growth factor(VEGF)and tumor necrosis factor(TNF)-αwere detected by immunoturbidimetry and enzyme-linked immunosorbent assay,respectively.Results Compared with the normal group,the levels of TC,TG,fasting blood glucose,VEGF,TNF-αand NO in model group were significantly increased(P<0.05);compared with the model group,the levels of TC,TG,fasting blood glucose,VEGF,TNF-αand NO in the down-regulated miR-184 group were significantly increased(P<0.05);compared with the down-regulated miR-184 group,the levels of TC,TG,fasting blood glucose,VEGF,TNF-αand NO in the up-regulated miR-184 group were significantly decreased(P<0.05);compared with the normal group,the level of GSH in the model group was significantly decreased(P<0.05);compared with the model group,the level of GSH in the down-regulated miR-184 group was significantly increased(P<0.05);compared with the down-regulated miR-184 group,the level of GSH in the up-regulated miR-184 group was significantly increased(P<0.05);compared with the normal group,the expression of glycogen synthase kinase(GSK)-3βwas significantly increased and the expressions ofβ-catenin(catenin)and proto-oncogene(C-Myc)were significantly decreased(P<0.05);compared with the model group,the expression of GSK-3βwas significantly increased and the expressions ofβ-catenin and C-Myc in the down-regu
