摘 要:目的运用代谢组学及生物效应网络方法研究人参皂苷Rg1-丹参素(GRg1-T)的保肝作用机制。方法将24只大鼠作为研究对象,采用简单随机化法将其分为空白组、模型组、GRg1-T组、联苯双酯(DDB)组,每组各6例。模型组给予CCl4油溶液。观察大鼠状态,测定生化指标;采用液相色谱-质谱联用技术(HPLC-MS)结合偏最小二乘辨别分析法,考察各组大鼠血清中的内源性代谢差异,筛选出肝损伤的生物标志物;将标志物代入到干预组中,寻找抗肝损伤潜在的生物标志物。结果模型组的谷丙转氨酶(ALT)、谷草转氨酶(AST)水平高于空白组,差异有统计学意义(P<0.001);GRg1-T和DDB组的ALT、AST水平低于模型组,差异有统计学意义(P<0.001)。鉴定出8种肝损伤的生物标志物;4种代谢物对肝损伤起到保护作用,分别为氨基丙酸、L-鸟氨酸、吡哆醛、卵磷脂。结论GRg1-T可能通过影响氨基酸、胆固醇的代谢及醛氧化酶的表达发挥肝保护作用。Objective To study the hepatoprotective mechanism of Ginsenoside Rg1-Tanshinol(GRg1-T)by metabonomics and bioeffect network.Methods A total of 24 rats were divided into blank group,model group,GRg1-T group and biphenyl diester(DDB)group by simple randomization,with 6 cases in each group.The model group was given CCl4 oil solution.The state of rats was observed and biochemical indexes were determined.Liquid chromatography-mass spectrometry(HPLC-MS)combined with partial least squares discrimination analysis was used to investigate the differences of endogenous metabolism in serum of each group of rats,and to screen out biomarkers of liver injury.The biomarkers were substituted into the intervention group to search for potential biomarkers against liver injury.Results The levels of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in model group were higher than those in blank group,the differences were statistically significant(P<0.001).ALT and AST levels in GRg1-T and DDB groups were lower than those in model group,and the differences were statistically significant(P<0.001).Eight biomarkers of liver injury were identified.Four metabolites,amino-propionate,L-ornithine,pyridoxal and lecithin,have protective effects on liver injury.Conclusion GRg1-T plays a protective role in liver by affecting the metabolism of amino acids,cholesterol and the expression of aldehyde oxidase.
